羌医制剂续筋接骨颗粒对大鼠骨缺损的修复作用及拆方研究

Repair Effect of Qiang Medical Prescription Xujin Jiegu Granule for Rat Bone Defect and Its Separated Prescrip-tion Study

目的:评估羌医制剂续筋接骨颗粒促进骨折愈合的药效,以及对处方进行拆方研究,发现关键药物组合。方法:采用大鼠股骨钻孔造成股缺损,灌胃给予续筋接骨颗粒浸膏(30、15、7.5g/kg),连续30天,取材后以组织学分级评分和骨痂厚度为指标评估骨缺损愈合程度;对续筋接骨处方按君臣药组和佐使药组进行拆方,比较全方及两组拆方对前述模型动物骨缺损愈合的影响。结果:续筋接骨颗粒浸膏各剂量组灌胃给予均可提高组织学分级评分、降低骨痂厚度;续筋接骨全方及君臣药组浸膏灌胃给予均可提高组织学分级评分、降低骨痂厚度,且全方组药效与君臣药组组间无显著性差异,而显著优于佐使药组。结论:续筋接骨颗粒在1/2临床等效剂量及以上剂量均可促进大鼠骨缺损愈合,提示在临床应用中可适当降低剂量以提高药物经济性、降低毒副反应;该方促进骨缺损愈合的药效主要与处方中君臣药组有关,提示可通过进一步筛选以确定关键药效物质,为新药研发及临床应用提供基础。

Objective:To study the promoting effect of Qiang medical prescription Xujinjiegu Granule on bone fracture healing,and discover the key drug combinations by separating the prescription.Methods:Rat bone defect model was established by bone drilling on femur.After modeling,Xujin Jiegu Extractum of different doses(30,15,7.5g/kg)were administrated orally for consistently 30days.After administration,rat shinbones were collected to evaluate the repair effect on bone defect by its histological score and the thickness of the callus.Xujin Jiegu Prescription were separated into Junchen drugs group and Zhuoshi drugs group,then repair effect of the original prescription and separated groups were compared.Results:In Xujin Jiegu Extractum groups,the histological scores were increased,the thickness of the callus was decreased.In Xujin Jiegu original group,Junchen drugs group and Zhuoshi drugs group,the histological scores were increased,the thickness of the callus was decreased.Compared with Zhuoshi drugs group,the original group improved the bone defect more significantly,while had no significant difference with Junchen drugs group.Conclusion:Xujin Jiegu Granule has repair effect on rat bone defect,on half dose of its clinical dosage and above,which suggests that the clinical dosage may be reduced to increase the economical efficiency and reduce its adverse reaction.Its bone repair effect may be related to the Junchen drugs in the prescription,which suggests that we can screen the key effective drugs for new drug research and clinical use.
