摘要
目的探讨颞叶内侧癫痫(MTLE)患者脑脊液中微小RNA-124(miR-124)和环磷酸腺苷反应元件结合蛋白(CREB)表达与认知功能关系。方法选取2016年6月至2019年6月本院神经内科收治的100例MTLE患者作为研究对象,选取同期接受脑脊液检查的80例无神经系统疾病、无癫痫史患者作为对照组,采用实时荧光定量PCR检测脑脊液中miR-124、CREB mRNA表达,观察两组患者miR-124、CREB mRNA表达及认知功能变化,分析miR-124、CREB mRNA表达相关性及与认知功能的关系。结果研究组患者脑脊液miR-124mRNA表达量明显低于对照组(P<0.05),CREB mRNA表达量明显高于对照组(P<0.05);研究组患者言语智商(VIQ)、操作智商(PIQ)、记忆商(MQ)评分均明显低于对照组(P<0.05);MTLE患者miR-124mRNA与CREB mRNA表达呈负相关(P<0.05);miR-124表达与VIQ、PIQ、MQ评分呈正相关(P<0.05),CREB表达与VIQ、PIQ、MQ评分呈负相关(P<0.05)。结论MTLE患者脑脊液miR-124低表达,CREB mRNA高表达,二者呈负相关,并与患者认知功能相关。
Objective To investigate the relationships between the expressions of microRNA-124(miR-124)and cyclic adenosine monophosphate response element binding protein(CREB)in cerebrospinal fluid and cognitive function in patients with medial temporal lobe epilepsy(MTLE).Methods A total of 100 patients with MTLE admitted to the Department of Neurology of our hospital from June 2016 to June 2019 were selected as the study subjects,and 80 patients without neurological diseases and epilepsy who underwent cerebrospinal fluid examination during the same period were selected as the control group.Real-time fluorescence quantitative PCR was used to detect the expressions of miR-124 and CREB mRNA in cerebrospinal fluid,the changes of the expressions of miR-124 and CREB mRNA in cerebrospinal fluid and cognitive function were observed,and the correlation between the expressions of miR-124 and CREB mRNA and their relationships with cognitive function were analyzed.Results The expression of miR-124 in cerebrospinal fluid of the study group was significantly lower than that of the control group(P<0.05),and the expression of CREB mRNA was significantly higher than that of the control group(P<0.05);the scores of verbal intelligence quotient(VIQ),performance intelligence quotient(PIQ)and memory quotient(MQ)in the study group were significantly lower than those in the control group(P<0.05);the expressions of miR-124 and CREB mRNA in MTLE patients were negatively correlated(P<0.05);the expression of miR-124 was positively correlated with VIQ,PIQ and MQ scores(P<0.05),while the expression of CREB was negatively correlated with VIQ,PIQ and MQ scores(P<0.05).Conclusion The expression of miR-124 in cerebrospinal fluid of patients with MTLE is low and CREB mRNA is high,they are negatively correlated,and are correlated with cognitive function of patients.
