摘要
目的探究miR-1247-3p在乳腺癌细胞中的表达及对乳腺癌细胞增殖、迁移及侵袭的影响。方法应用qPCR检测miR-1247-3p在乳腺癌细胞及乳腺上皮细胞中的表达水平;合成miR-1247-3p的mimic、inhibitor、NC,并转染至乳腺癌细胞;CCK8法、细胞划痕、Transwell小室法检测转染后乳腺癌细胞的增殖、迁移和侵袭能力;采用生物信息学的方法预测miR-1247-3p的靶基因,荧光素酶报告实验验证靶基因,Western blot检测转染miR-1247-3p mimic、inhibitor、NC后靶基因的表达水平;转染靶基因的siRNA后,检测乳腺癌细胞的增殖、迁移和侵袭能力。结果miR-1247-3p在乳腺癌细胞中表达量显著上调,且miR-1247-3p能够在体外促进乳腺癌细胞的增殖、划痕的愈合以及侵袭能力;预测并验证B4GALT3为miR-1247-3p的靶基因,miR-1247-3p能够下调B4GALT3的表达;siRNA下调B4GALT3的表达能够促进乳腺癌细胞的增殖、迁移和侵袭能力。结论miR-1247-3p通过下调靶基因B4GALT3的表达而促进乳腺癌细胞的增殖、迁移以及侵袭。
Objective To investigate the expression of miR-1247-3p in breast cancer cells and its effect on proliferation,migration and invasion of breast cancer cells.Methods qPCR was used to detect the expression level of miR-1247-5p in breast cancer cells and mammary epithelial cells.The mimic,inhibitor and NC of miR-1247-3p were synthesized and transfected into breast cancer cells,CCK8 method,cell scratches,Transwell chamber method were used to detect the ability of proliferation,migration and invasion of breast cancer cells after transfection.The target gene of miR-1247-3p was predicted by bioinformatics method,and the target gene was verified by luciferase reporter assay.Western blot was used to detect the expression level of the target gene;after transfecting the siRNA of the target gene,the proliferation,migration and invasion ability of the breast cancer cells are detected.Results The expression of miR-1247-3p was up-regulated in breast cancer cells,and miR-1247-3p could promote the proliferation,scratch healing and invasion of breast cancer cells in vitro.Prediction and verification of B4GALT3 was miR-1247-3p target gene,and miR-1247-3p can down-regulate the expression of B4GALT3;down-regulation of B4GALT3 by siRNA can promote the proliferation,migration and invasion of breast cancer cells.Conclusion miR-1247-3p promotes the proliferation,migration and invasion of breast cancer cells by down-regulating the expression of the target gene B4GALT3.
