‘镇研958六号’辣椒杂交种纯度的SRAP鉴定 被引量:1

摘要 以辣椒品种‘镇研958六号’及其父母本为材料,采用相关序列扩增多态性(SRAP)分子标记方法,建立基于DNA分子标记的‘镇研958六号’辣椒品种纯度鉴定方法,以期为提高大田制种纯度提供分子辅助工具。结果发现,从30对SRAP标准引物中筛选出引物E3M1能区分‘镇研958六号’及其亲本;应用此引物对‘镇研958六号’杂交种进行纯度鉴定,其纯度结果为98%,与田间鉴定结果基本一致。结果表明,引物E3M1可以作为鉴定‘镇研958六号’纯度的分子标记,SRAP分子标记用于辣椒杂交种子纯度鉴定是可行的。 DNA molecular markers were used for testing the seed purity of F1 hybrid hot pepper cultivar‘Zhenyan 958 No.6’with the aim to establish a rapid and reliable method for genetic purity testing of commercial hybrid pepper seeds.Genetic relationships of‘Zhenyan 958 No.6’and its parents were analyzed with 30 sequence-related amplified polymorphism(SRAP)primers.Among these primers,one primer(E3M1)amplifying both female and male-parent specific products simultaneously,was selected for the genetic purity test.A total of 49‘Zhenyan 958 No.6’hybrid individuals were analyzed with the selected primer.The results showed that the overall genetic purity of the hybrid seeds was about 98%,which was generally consistent with morphological analysis in field experiment.SRAP marker identified in this study is a good option for establishment of the seed quality control system in commercial seed production of pepper.
