酶解辅助乙醇回流提取欧李中有机酸 被引量:3

Organic Acids Extracted from Cerasus Humilis with Ethanol Hot Reflux Assisted by Enzymatic Treatment

摘要 以欧李作为原料,用果胶酶进行酶解处理,然后通过乙醇回流法提取欧李中的有机酸。文章探讨了酶解反应处理的温度、加酶量、反应时间的因素对有机酸提取率的影响,并进行了正交试验优化。结果表明,欧李中有机酸的最佳酶处理工艺条件为加酶量0.5%、酶解温度60℃、酶解时间1 h;在此条件下,欧李中有机酸提取率为89.94%。HPLC分析得出,有机酸组成以苹果酸为主,含量为21.574 mg/mL。研究结果表明:通过酶解反应处理辅助乙醇回流,适合从欧李中提取有机酸。此法所得的有机酸以苹果酸为主,提取率高、工艺简单,易于工业化。 Pectinase was used to pretreat Cerasus humilis,and then the organic acids in Cerasus humilis were extracted by ethanol hot reflux.The effects of 3 factors including enzyme reaction temperature,reaction time and enzyme addition on the extraction rate of organic acids were investigated.By orthogonal test analysis,optimum extraction process were as follows:enzyme reaction temperature of 60℃,reaction time of 1 h,and enzyme addition of 0.5%.Under the optimal conditions,the organic acid extraction rate of Cerasus humilis was 89.94%,based on the theoretical content.Malic acid of 21.574 mg/mL is the main organic acid by HPLC.This results indicates the process is simple,feasible,and suitable for the extraction of ethanol hot reflux by enzymatic pretreatment,and the extract of organic acid mainly is malic acid.
