甘草次酸对人脑胶质瘤细胞U251增殖的抑制作用 被引量:2

摘要 目的研究甘草次酸对人脑胶质瘤细胞U251增殖的影响。方法将人脑胶质瘤细胞U251分为5组:空白对照组、阳性对照组和低、中、高3个浓度实验组。空白对照组不加任何药物,阳性对照组加入白藜芦醇40μmol·L^-1,低、中、高3个浓度实验组加入甘草次酸50,100,200μmol·L^-1。通过甲基四氮唑蓝检测细胞的增殖率,以Annexin V/PI双标记流式细胞术检测人脑胶质瘤细胞U251的凋亡比例,以蛋白质印迹法检测检核转录因子κB(NF-κB)蛋白相对表达量。结果孵育48 h后,空白对照组和低、中、高3个浓度实验组的细胞增殖率分别为(104.45±3.56)%,(80.56±3.98)%,(72.11±4.67)%和(56.56±4.51)%;这4组的凋亡细胞比例分别为(14.27±1.45)%,(27.78±3.35)%,(37.85±3.12)%和(59.12±4.17)%;这4组的NF-κB蛋白的相对表达量分别为3.18±0.22,2.11±0.21,1.36±0.13和1.15±0.19。上述指标:低、中、高3个浓度实验组与空白对照组相比,差异均有统计学意义(均P<0.05)。结论甘草次酸通过影响NF-κB信号通路抑制人脑胶质瘤细胞U251增殖。 Objective To study the effect of glycyrrhetinic acid on Glioma cell line U251.Methods Glioma cells U251 were divided into five groups:Blank control group(was not any treatment),positive control group(40μmol·L^-1 resveratrol),and experimental-L,experimental-M,experimental-H groups(50,100,200μmol·L^-1 glycyrrhetinic acid).The proliferation rate of Glioma cells U251 was measured by methyl thiazolyl tetrazolium.The proportion of apoptosis was detected by cytometry with Annexin V/PI.The relative expression of nuclear factor-κB(NF-κB)protein was detected by Western blotting.Results After incubation for 48 h,proliferation rates in blank control group,and experimental-L,experimental-M,experimental-H groups were(104.45±3.56)%,(80.56±3.98)%,(72.11±4.67)% and(56.56±4.51)%;proportions of apoptosis in the four groups were(14.27±1.45)%,(27.78±3.35)%,(37.85±3.12)%and(59.12±4.17)%;relative expression levels of NF-κB protein in the four groups were 3.18±0.22,2.11±0.21,1.36±0.13 and 1.15±0.19.Compared between three concentrations experimental groups and blank control group,the differences of the factors were significant(all P<0.05).Conclusion Glycyrrhetinic acid inhibits proliferation of Glioma cell line U251 by inhibiting NF-κB signaling pathway.
