摘要
青光眼是导致视力不可逆损伤的第一大原因,对于一些药物无法控制的青光眼中小梁切除术是最常用的治疗青光眼的方法。但是术后巩膜瓣瘢痕化和结膜组织的纤维化等使滤过道阻塞的原因均会导致青光眼滤过手术失败。环状RNA(circRNA)是一类具有闭合环状结构的RNA,且稳定表达于多种生物组织细胞中,曾一度被认为是基因转录过程的中间产物、副产物以及一些偶然的剪接错误而产生的。随着各种测序技术的精细化及生物信息分析方法的不断改进,circRNA潜在的功能也逐渐被发觉,这也使得circRNA得到了广泛的关注。circRNA可通过多种途径调控基因的表达,在组织纤维化方面也有非常重要的作用。本文将从青光眼滤过手术失败原因,组织纤维化中各种机制及circRNA在各个器官组织中的作用以及它们之间的联系方面进行综述,希望能够为青光眼滤过手术后抗纤维化提供新的研究及治疗方向。
Glaucoma is the main cause of irreversible visual impairment,and trabeculectomy is the most commonly used treatment for glaucoma when intraocular pressure can not be controlled by medication.However,the causes of filtering passage obstruction,such as scarring of scleral flap and fibrosis of conjunctival tissue,can lead to failure of the filtering surgery.Circular RNA(circRNA)is a kind of RNA with a special closed circular structure,which is stably expressed in a variety of biological tissues and cells.They were once considered as intermediate products,by-products or results of accidental splicing errors during the genetic transcription process.With the development and improvement of high-throughput sequencing technology and bioinformatics analysis methods,the potential function of circRNAs have been gradually discovered,and thus cirRNAs received widespread attention.The circRNAs can regulate gene expression in many ways,especially in tissue fibrosis.This article reviews the reasons of failure of glaucoma filtering surgery,various mechanisms of tissue fibrosis and the role of circRNA in various organs and tissues,hoping to provide a new research and treatment direction for anti-fibrosis after glaucoma filtering surgery.
