摘要
细胞计数目前最精准的计数方法为流式计数,其原理是先将细胞聚焦成线,使其单个通过检测位,实现细胞绝对计数。现有的细胞聚焦方式为鞘液聚焦,需要复杂的进液系统。因此,利用惯性聚焦替代鞘液聚焦,设计合理的正弦型惯性聚焦流道,通过惯性力与迪恩拽力将粒子进行汇聚。利用聚苯乙烯微球模拟细胞,进行流体聚焦实验。实验结果表明:设计的惯性聚焦流道在550μL/min的流速下,能实现7μm、10μm、12μm混合微球的惯性聚焦。
Cell counting is currently the most accurate counting method by flow counting.The principle is to first focus the cells into a line so that they individually pass through the detection position to achieve absolute cell counting.The existing cell focusing method requires a complex liquid feeding system for sheath fluid focusing.Using inertial focusing instead of sheath focusing,a reasonable sinusoidal inertial focusing channel was designed,and particles were converged by inertial force and Dean drag force.Polystyrene microspheres were used to simulate cells and fluid focusing experiments were performed.Experiment results show that the designed inertial focusing channel can achieve inertial focusing of 7μm,10μm,12μm hybrid microspheres at a flow rate of 550μL/min.
