摘要
目的:建立反相高效液相色谱法测定民族药材蚤状车前子中京尼平苷酸和毛蕊花糖苷的含量。方法:选用ZORBAX Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.2%磷酸溶液为流动相进行梯度洗脱,检测波长240nm,流速1.0 ml·min^-1,柱温35℃。结果:京尼平苷酸和毛蕊花糖苷分别在0.0151~0.6048 mg·ml^-1和0.0051~0.2053 mg·ml^-1范围内线性关系良好(r=0.9999),精密度、稳定性、重复性、回收率试验RSD均小于2%,平均加样回收率分别为101.1%和102.8%,RSD分别为1.27%,0.94%(n=6)。结论:该方法简便准确、灵敏度高、重现性好,可作为蚤状车前子药材中京尼平苷酸和毛蕊花糖苷的含量测定方法。
Objective: To establish an HPLC method to determine the contents of geniposidic acid and verbascoside in Plantago ovata Forssk. Methods: The separation was performed on a ZORBAX Eclipse XDB-C18 column( 250 mm × 4.6 mm,5 μm). Gradient elution was applied with acetonitrile-0.2% phosphoric acid solution as the mobile phase at a flow rate of 1.0 ml·min^-1. The detection wavelength was 240 nm,and the column temperature was 35 ℃ . Results: The calibration curve of geniposidic acid and verbascoside was linear within the range of 0.015 1-0.604 8 mg·ml^-1 and 0.005 1-0.205 3 mg·ml^-1( r =0.999 9),respectively. The RSDs of precision,solution stability,repeatability and recovery were all less than 2%,and the average recovery was 101.1%( RSD = 1.27%) and 102.8%( RSD = 0.94%)( n = 6),respectively. Conclusion: The method is simple,accurate,sensitive and reproducible,and can be used for the quality control of Plantago ovata Forssk.
