柱前衍生法测定烟叶中的7种生物胺

Determination of 7 biogenic amines in tobacco leaves by a pre-column derivatization method

建立了丹磺酰氯柱前衍生高效液相色谱法(HPLC)测定烟草基因编辑素材中的7种生物胺的方法。采用0.4 mol/L HClO4进行样品超声提取,提取液经过pH调整后,利用丹磺酰氯(4 mg/mL)进行柱前衍生;衍生后的样品溶液利用优化的HPLC-DAD色谱方法进行检测;方法在1.0~50μg/mL范围内具有较好的线性(R^2>0.98),检出限在0.01~0.15μg/mL之间,定量限在0.05~0.25μg/mL之间,日间和日内精密度均小于5%,不同加标水平下,平均回收率在86.7%~124.0%之间。

A method for the determination of 7 biogenic amines in tobacco genetic material by sulfonyl chloride pre-column derivatization high performance liquid chromatography(HPLC)was developed.Ultrasonic extraction were carried out with 0.4 mol/L HClO4.After pH adjustment,the extract was pre-column derivatized with dansyl chloride(4 mg/m L).The sample solution after derivatization was detected by optimized HPLC-DAD chromatography.The method had good linearity(R^2>0.98)in the range of 1.0-50μg/m L,the detection limits were between 0.01-0.15μg/m L and the limits of quantification were between 0.05 and 0.25μg/m L.The precision was less than 5% in interday and intraday.Under different spiked levels,the average recoveries were between 86.7% and 124.0%.
