单采血小板血浆低密度脂蛋白在保存期间对血小板免疫活化的影响 被引量:1

摘要 目的:探讨单采血小板血浆低密度脂蛋白(low-density lipoprotein,LDL)升高是否对血小板免疫活化产生影响。方法:选取符合国家献血标准的单采血小板献血员血小板标本35例,于血小板保存箱(22℃±2℃)振摇保存,其中LDL正常组(LDL<3.4 mmol/L)30例,LDL升高组(LDL≥3.4 mmol/L)5例,流式细胞术检测血小板表面活化指标CD63、CD36、CD40以及CD62p表达水平,比较不同LDL水平对单采血小板免疫活化的影响。结果:随着保存时间的延长,血小板CD63、CD36、CD40以及CD62p表达水平无显著改变(P>0.05),不同保存时间LDL升高组与LDL正常组比较,CD63、CD36、CD40以及CD62p表达水平的差异无统计学意义(P>0.05)。保存第2、3、4、5天的血小板经室温不同静置时间,LDL升高组与LDL正常组比较,CD63、CD36、CD40以及CD62p表达水平的差异均无统计学意义(P>0.05)。结论:健康献血者单采血小板血浆LDL水平对血小板保存期间的免疫活化无显著影响,LDL水平较高的单采血小板不会激发血小板免疫活化,从而不影响临床输注效果。 Objective:To investigate the effect of plasma low-density lipoprotein(LDL)on activation during apheresis platelets storage.Methods:Collection in accordance with the national standard of the blood donation for 35 cases apheresis platelets,including 30 cases in the LDL normal group(LDL<3.4 mmol/L)and 5 cases in the LDL increased group(LDL≥3.4 mmol/L),were stored in the platelet preservation box(22±2℃)with shaking.Flow cytometry was used to detect the expression levels of CD63,CD36,CD40 and CD62p on the surface of platelets,and to compare the effects of different LDL levels on the immune activation of apheresis platelets.Results:With the increase of storage time,there was no significant difference in the expression levels of CD63,CD36,CD40 and CD62p in apheresis platelets(P>0.05),and there was also no statistical difference between the LDL increased group and the LDL normal group in different storage time(P>0.05).In addition,there was no statistically significant difference in the expression of CD63,CD36,CD40,and CD62p between the platelets stored at 2,3,4,and 5 days at room temperature for different standing time in the LDL increased group and the LDL normal group(P>0.05).Conclusion:The plasma LDL level of apheresis platelets in healthy blood donors had no significant effect on the immune activation of platelets during in vitro preservation.Apheresis platelets with high LDL levels could not effectively stimulate the immune activation of platelets,so the clinical platelet infusion effect would not be affected.
