华蟾素对卵巢癌细胞增殖侵袭能力的抑制作用 被引量:4

摘要 目的探讨华蟾素对卵巢癌SKOV-3细胞系增殖能力及侵袭能力的影响及其作用机制。方法将对照组,低、中、高剂量组SKOV-3细胞分别加入含有质量浓度为0,0.1,1.0,10.0 mg/L华蟾素的培养液,培养24,48,72 h后,检测其增殖抑制率、凋亡率和细胞侵袭能力,以及SKOV-3细胞Bcl-2,Bax,E-cadherin和基质金属蛋白酶-7(MMP-7)蛋白的表达情况。结果经不同质量浓度华蟾素处理不同时间后,SKOV-3细胞侵袭力随着药物质量浓度的升高和处理时间的延长而显著降低,增殖抑制率和凋亡率显著升高;细胞E-cadherin及Bax蛋白表达量随着药物质量浓度的升高和处理时间的延长而显著升高,MMP-7和Bcl-2蛋白表达量随药物质量浓度的升高和处理时间的延长而显著降低。结论华蟾素可通过调节卵巢癌细胞的凋亡能力而抑制其增殖能力和侵袭能力。 Objective To investigate the effect and mechanism of cinobufacini on the proliferation and invasion of oophoroma SKOV-3 cells.Methods SKOV-3 cells were cultured in medium containing 0,0.1,1,10 mg/L cinobufacini for 24,48,and 72 h respectively.The proliferation inhibition rate,apoptotic rate and invasive ability of SKOV-3 cells were detected,and the expression of Bcl-2,Bax,E-cadherin and matrix metalloenzyme-7(MMP-7)protein was also detected.Results The invasiveness of SKOV-3 cells decreased significantly with the increase of drug concentration and treatment time,and the proliferation inhibition rate and apoptosis rate increased significantly with the increase of drug concentration and treatment time.The expression of E-cadherin and Bax increased significantly with the increase of drug concentration and treatment time,while the expression of MMP-7 and Bcl-2 decreased significantly with the increase of drug concentration and treatment time.Conclusion Cinobufacini can inhibit the proliferation and invasiveness of oophoroma cells by regulating the apoptotic ability of oophoroma cells.
