摘要
目的建立同时测定鸡骨草中不同部位相思子碱和夏佛塔苷2种活性成分含量的超高效液相色谱(UPLC)法。方法色谱柱为HSS T3 C18柱(100 mm×2.1 mm,1.8μm),流动相为乙腈(A)-0.1%甲酸(B),梯度洗脱,流速为0.3 m L/min,检测波长为270 nm,柱温为30℃,进样量为2μL。结果相思子碱和夏佛塔苷的质量浓度线性范围分别为5.068~126.7μg/m L(r=0.9999)和9.602~240.1μg/m L(r=1.0000);平均回收率分别为98.38%和97.39%,RSD分别为1.30%和1.02%(n=9);20批不同部位样品中相思子碱和夏佛塔苷的含量范围分别为0.1197~0.2651 mg/g和0.2011~6.0421 mg/g。结论该方法简便、专属性强,增加了质量控制指标,可为该饮片质量标准的完善提供依据。
Objective To establish a method for the simultaneous determination of abrine and schaftoside in Abri herba from different medicinal portions by ultra performance liquid chromatography(UPLC).Methods The separation was performed by HSS T3 C18 column(100 mm×2.1 mm,1.8μm)with the mobile phase consisting of acetonitrile(A)-0.1%formic acid(B)by gradient elution at a flow rate of 0.3 m L/min;the wavelength was 270 nm,the column temperature was 30℃,and the injection volume was 2μL.Results The linear ranges of abrine and schaftoside were 5.068-126.7μg/m L(r=0.9999)and 9.602-240.1μg/m L(r=1.0000).The average recoveries were 98.38%and 97.39%,respectively,and RSDs were=1.30%and 1.02%(n=9).The content ranges of abrine and schaftoside in 20 batches of sample were 0.1197-0.2651 mg/g and 0.2011-6.0421 mg/g.Conclusion This method is simple with good specificity and increases the quality control index,which can provide reference for the improvement of the intrinsic quality of Abri herba.
