公猪睾丸、附睾及精子中硒蛋白编码基因相对表达量分析 被引量:1

摘要 【目的】考察公猪生殖系统中硒蛋白编码基因表达情况,初步筛选与其繁殖性能密切相关的硒蛋白编码基因。【方法】采用RT-qPCR技术对成年和半月龄DLY三元杂交公猪的睾丸、附睾及杜洛克公猪的精子进行25个硒蛋白编码基因表达量分析,采用2^(-△△Ct)法计算各基因相对表达量。【结果】①GPX3和TXNRD2在仔公猪睾丸中显著高表达(P<0.05),GPX6、SELENOM和SELENON在仔公猪附睾中显著高表达(P<0.05);②GPX4、SELENOH、SELENOK、SELENOS、SELENOT、SELENOV、SEPHS2、TXNRD1等8个硒蛋白编码基因在成年公猪睾丸中表达量显著高于其余样品中表达量(P<0.05);③MSRB1和SELENOI在成年公猪附睾中表达量显著高于仔公猪附睾中表达量(P<0.05)。【结论】在公猪生殖系统中,13个硒蛋白编码基因表达具有组织特异性,2个硒蛋白编码基因表达受性成熟的影响,揭示这些差异表达的硒蛋白编码基因与相应器官生殖功能密切相关。 【Objective】The purpose of this study is to investigate the mRNA abundance of selenoprotein encoding genes in the reproductive system of boar and screen the key selenoprotein encoding genes related to male fertility.【Method】The m RNA abundance of 25 selenoprotein coding genes in the testes and the epididymides of adult and half-month-old DLY boar and Duroc boar spermium were determined with RT-qPCR.The 2-△△Ct method was utilized to calculate the relative mRNA levels of each target gene.【Result】It was found that:①GPX3 and TXNRD2 significantly high expressed in the testes of half-amonth-old piglets,while GPX6,SELENOM and SELENON significantly high expressed in the epididymides of half-a-month-old piglets(P<0.05);②Eight selenoprotein coding genes,namely GPX4,SELENOH,SELENOK,SELENOS,SELENOT,SELENOV,SEPHS2,and TXNRD1,significantly high expressed in the testes of adult boars(P<0.05);③The relative mRNA levels of MSRB1 and SELENOI in the epididymides of adult boars were significantly higher than that of male piglets(P<0.05).【Conclusion】In the boar reproductive system,13 selenoprotein coding genes were tissue-specific expression,2 selenoprotein coding genes were influenced by the age.It reveals that the selenoprotein coding genes with differentially expression are closely related to the reproductive functions of corresponding organs.
