摘要
目的探讨幽门螺杆菌相关胃疾病患者胃黏膜组织中微小RNA-26a(miR-26a)、环氧化酶-2(COX-2)的表达及临床意义。方法收集本院2016年2月至2019年3月,幽门螺杆菌相关胃疾病患者95例,同期健康对照者30例,实时荧光定量PCR法检测胃黏膜活检组织中miR-26a、COX-2 mRNA表达水平,免疫组化法检测胃黏膜活检组织中COX-2的蛋白表达。结果与健康对照比较,慢性浅表性胃炎、慢性萎缩性胃炎、肠上皮化生、胃癌患者胃黏膜活检组织中miR-26a表达量依次降低(P<0.05),COX-2 mRNA、COX-2蛋白表达量依次升高(P<0.05)。胃癌黏膜活检组织中miR-26a、COX-2蛋白表达与分化程度和淋巴结转移相关(P<0.05)。胃癌黏膜活检组织中miR-26a和COX-2表达呈负相关(r=-0.807,P<0.05)。结论随幽门螺杆菌相关胃病进展,患者胃黏膜组织中miR-26a表达显著降低,COX-2蛋白表达显著升高,二者表达呈负相关,与幽门螺杆菌相关胃疾病发生发展密切相关。
Objective To investigate the expressions and clinical significances of microRNA-26a(miR-26a)and cyclooxygenase-2(COX-2)in gastric mucosa of patients with Helicobacter pylori related gastric diseases.Methods From February,2016 to March,2019,95 patients with Helicobacter pylori related gastric diseases and 30 healthy controls were collected for the study.The expression levels of miR-26a and COX-2 mRNA in gastric mucosa biopsies of these subjects were detected by real-time fluorescence quantitative PCR,and the expression of COX-2 protein was detected by immunohistochemistry.Results Compared with the healthy control group,the expression of miR-26a in gastric mucosa biopsies of patients with chronic superficial gastritis,chronic atrophic gastritis,intestinal metaplasia and gastric cancer decreased in turn(P<0.05),while the expressions of COX-2 mRNA and COX-2 protein increased in turn(P<0.05).The expressions of miR-26a and COX-2 proteins in gastric mucosa biopsies were correlated with the degree of differentiation and lymph node metastasis(P<0.05).There was a negative correlation between the expressions of miR-26a and COX-2 in gastric mucosa biopsies(r=-0.807,P<0.05).Conclusion With the development of Helicobacter pylori related gastropathy,the expression of miR-26a in gastric mucosa is decreased significantly,and the expression of COX-2 protein is increased significantly,during which they are negatively correlated,and also closely related to the occurrence and development of Helicobacter pylori related gastric diseases.
