重楼皂苷Ⅶ抑制PC3细胞的增殖和周期阻滞的研究

Effects of Polyphyllin Ⅶ on the Cell Cycle of Human Prostate Cancer PC3 Cells and the Experiment Research

探讨重楼皂苷Ⅶ(PolyphyllinⅦ,PP7)对人前列腺癌PC3细胞增殖和周期的影响及其机制.采用Cell Counting Kit-8(CCk-8)方法观察PP7对PC3细胞增殖的影响.流式细胞术研究PP7对PC3细胞周期的作用.培养PC3细胞并给予0.33和0.5μg/mL的PP7.经过24 h后,采用Western blot法检测周期相关蛋白Cyclin D1和p21的表达变化.结果表明,PP7可明显抑制PC3细胞增殖,并呈明显的浓度依赖性,其IC50值为(0.67±0.02)μg/mL;PP7可上调PC3细胞p21的表达,下调Cyclin D1的表达.PP7将PC3细胞周期阻滞于G1期是通过上调p21,抑制Cyclin D1周期调控蛋白的表达实现的.

To investigate the effects of Polyphyllin Ⅶ(PP7)on the cell cycle of human prostate cancer PC3 cells and the potential mechanisms,the effects of different doses of PP7 on the proliferation of PC3 cells were observed by Cell Counting Kit-8(CCk-8)assay,and the effects of PP7 on the cell cycle of PC3 cells were investigated by flow cytometric analysis.After treated with different concentration(0.33,0.5μg/mL)of PP7,Western blot was used to test the expressions of the cycle associated protein CyclinD1 and p21 in PC3 cells.The results show that PP7 could obviously inhibit the proliferation of PC3 cells significantly in a dose dependent manner,and the IC50 was(0.67±0.02)μg/mL.PP7 could reduce the expression of Cyclin D1,while increased the expression of p21 in PC3 cells.PP7 can arrest the cell Cyclin G1 phase by increasing the expression of p21,while decreasing the expression of Cyclin D1 in PC3 cells.
