18ka转运蛋白配体对抑郁大鼠不同脑区四氢孕酮水平影响的研究

Effects of 18ka Transporter(TSPO)on the Content of Allopregnanolone in Different Brain Regions of Depressed Rats

摘要 目的研究18ka转运蛋白配体对抑郁大鼠不同脑区四氢孕酮水平的影响。方法应用大鼠慢性不可预知温和应激抑郁模型(CUMS),通过灌胃18ka转运蛋白配体AC-5216,进行采用酶联免疫吸附试验检测大鼠不同脑区四氢孕酮的含量,评价AC-5216对抑郁大鼠不同脑区四氢孕酮水平影响。结果大鼠强迫游泳和蔗糖偏好实验证明CUMS造模成功。给予AC-5216后,大鼠前额皮质层、海马体、杏仁核脑区的四氢孕酮含量显著升高;前部扣带回、腹中央纹状体四氢孕酮含量无显著性差异。结论AC-5216的抗抑郁作用与前额皮质层、海马体、杏仁核内四氢孕酮水平相关。 OBJECTIVE To investigate the effects of 18ka transporter ligand on the content of allopregnanolone in different brain regions of depressed rats.METHODS TSPO ligand AC-5216 was administered to rats by chronic unpredictable mild stress depression model(CUMS).Enzyme-linked immunosorbent assay(ELISA)was used to detect the content of allopregnanolone in different brain regions of rats.RESULTS CUMS model was successfully constructed by forced swimming test and sucrose preference test in rats.After the administration of AC-5216,the content of allopregnanolone in the prefrontal cortex,hippocampus and amygdala was significantly increased.There was no significant difference in anterior cingulate gyrus and ventral central striatum.CONCLUSION The antidepressant-like effect of AC-5216 is correlated with the content of allopregnanolone in the prefrontal cortex,hippocampus and amygdala.
