miR-142-3p对子宫内膜癌细胞增殖、迁移及侵袭的影响 被引量:1

摘要 目的探讨miR-142-3p对子宫内膜癌细胞增殖、迁移和侵袭的影响。方法采用qRT-PCR检测子宫内膜癌细胞(Ishikawa和HEC-1A)和正常子宫内膜细胞(HEC-251)中miR-142-3p的表达水平;采用MTT和Transwell法评估miR-142-3p高表达对子宫内膜癌细胞增殖、迁移以及侵袭能力的影响;采用Western blot法测定迁移侵袭相关蛋白的表达水平。结果与HEC-251细胞相比,Ishikawa和HEC-1A细胞中miR-142-3p表达水平显著降低(P<0.05);与Control组以及miR-NC组相比,miR-142-3p高表达显著抑制HEC-1A细胞增殖、迁移及侵袭(P<0.05)。此外,miR-142-3p高表达显著抑制MMP-2和MMP-9的表达(P<0.05)。结论miR-142-3p能够抑制子宫内膜癌细胞增殖、迁移及侵袭能力。 Objective To explore the effect of mi R-142-3p on the proliferation,migration and invasion of endometrial cancer cells.Methods The expression of mi R-142-3p in human endometrial cancer cell lines(Ishikawa and HEC-1A)and normal endometrial cell line(HEC-251)was measured by q RT-PCR.MTT assay and transwell assay were used to detect the proliferation,migration and invasion of HEC-1A cells.Western blot assay was used to measure the expression of MMP-2 and MMP-9 in HEC-1A cells.Results When compared with HEC-251 cells,the expression of mi R-142-3p was significantly lower in Ishikawa and HEC-1A cells(P<0.05).mi R-142-3p overexpression markedly inhibit the proliferation,migration and invasion of HEC-1A cells(P<0.05).Moreover,mi R-142-3p overexpression significantly represses the expression of MMP-2 and MMP-9 in HEC-1A cells(P<0.05).Conclusion mi R-142-3p could inhibit the proliferation,migration and invasion of endometrial cancer cells.
