摘要
目的优选紫花地丁中秦皮乙素、总香豆素的大孔树脂纯化工艺条件。方法以秦皮乙素、总香豆素为评价指标,通过7种大孔树脂(AB-8、D101、HPD-100、HPD-450、HPD-750、DM130、HPD-600)的静态吸附、动态解吸试验,确定HPD-100为最佳树脂,经L9(34)正交及单因素试验进行纯化工艺研究。结果最佳纯化条件为HPD-100大孔树脂,上样液pH值为5.0、柱径高比为1∶9、上样液浓度为1.0 g/g、上样体积流量为0.5 BV/h、洗涤用水量为4 BV、解吸溶剂为4 BV 50%乙醇、洗脱体积流速为2 BV/h,纯化后,秦皮乙素、总香豆素含量分别提高5.4倍、5.5倍。结论HPD-100大孔树脂分离纯化紫花地丁效果良好,工艺稳定可行。
Objective To optimize the purification process of esculetin and total coumarin from Viola yedoensis by macroporous absorption resin.Methods Taking esculetin and total coumarin as evaluation indexes,HPD-100 is determined to be the best resin through the adsorption and desorption tests of seven macroporous resins(AB-8,D101,HPD-100,HPD-450,HPD-750,DM130,HPD-600).The purification process is determined by orthogonal and single factor tests.Results HPD-100 type macroporous absorption resin is the best choice for the purification of esculetin and total coumarin from Viola yedoensis,the optimized parameters are as follows:Drug concentration is 1.0 g/g with pH 5.0 and the ratio of diameter to height is 1:9 at a flow rate of 0.5 BV/h,4 BV of purified water is used to remove impurities,and then 2 BV 50%ethanol and 4 BV 50%ethanol are used as eluent at a flow rate of 2 BV/h.After purification,the content of esculetin and total coumarin increased 5.4 and 5.5 times,respectively.Conclusion The purification process of Viola yedoensis by HPD-100 macroporous resin is stable and feasible.
