复合酶法制备玉米活性肽及其功能特性的研究 被引量:1

摘要 采用碱性蛋白酶和胃蛋白酶制备玉米活性肽,以水解度为指标,分别对酶解温度、酶解时间、加酶量及pH值4个因素进行单因素试验及正交试验分析,确定双酶水解的最佳酶解条件。结果表明,碱性蛋白酶水解最佳条件为酶解温度60℃,酶解时间为1.5 h,加酶量为0.128 g,pH值为11,水解度为13.63%;胃蛋白酶水解最佳条件为酶解温度60℃,酶解时间为1.5 h,加酶量为0.008 g,pH值为2.1,水解度为22.12%;双酶水解后水解度可达29.88%;玉米肽对自由基清除率达25.62%。 In this study,the bioactive peptides were derived from corn gluten using alkaline protease and pepsin.The influence of hydrolysis temperature,hydrolysis time,enzyme concentration and pH value on degree of hydrolysis were studied by single factor and orthogonal experiments analysis.And then the optimum enzymatic hydrolysis condition was determined.The results showed that the optimum enzymatic conditions of alkaline protease were temperature 60℃,time 1.5 h,enzyme concentration 0.128 g,pH value 11,under optimum enzymatic hydrolysis conditions,the hydrolysis degrees was 13.63%.The best enzymatic conditions of pepsin were temperature 60℃,time 1.5 h,added enzyme volume 0.008 g,pH value 2.1,under optimum enzymatic hydrolysis conditions,the hydrolysis degrees was 22.12%.The hydrolysis degrees of double enzyme hydrolysis was up to 29.88%.The rate of free radical scavenging rate by corn peptide was up to 25.62%.
