摘要
为寻找效果好、可随时取用、占用空间小、步骤易操作的藻种保藏方法,以10株节旋藻/螺旋藻为试验对象进行耐低温试验,选择较耐低温螺旋藻F-1070为对象进行正交试验,探索以海藻糖为保护剂的浓缩藻液低温弱光保藏法的保藏效果。藻种的活力评价基于SLogistic模型对保藏藻种再生长曲线的拟合及对生长曲线一阶求导获得生长速率曲线,通过对两曲线相关指标的统计分析得到评价藻种活力的特征指标,用以创建浓缩藻液低温弱光保藏海藻糖法的评价体系,同时初步获得了微藻的保藏条件:弱光照下藻液浓度2×10^6CFU/mL^4×10^6CFU/mL、海藻糖浓度15%。
This research regarded Spirulina platensis F-1070 as experimental subjects,exploring an effectual and convenient microalgae preservation method that was able to have acquisition timely,less space required and simple operating steps. F-1070 was enduring low temperature obtained by single factor experimental way of 10 strains of algae. At the same time,the optimal concentration range of trehalose was determined. The storage conditions of preservation method were further optimized by orthogonal test. The experiment design was based on regrowth curve of preservation microalgae fitting by SLogistic regression models. The growth rate curve of microalgae was gained by the first-order derivation of the growth curve. Through the statistical analysis of both curves,this method got some characteristic indexes to estimate the microalgae vitality and evaluate the effect of preservation of microalgae concentrate in order to obtain the best conditions of the dim light low-temperature preservation method. And the preservation conditions of the obtained microalgae were 2×10^6 CFU/mL-4×10^6 CFU/mL of algae liquid and 15 % trehalose in low light.
