摘要 目的:观察龙眼叶总黄酮(total flavonoids of longan leaves,TFLL)对大鼠动脉粥样硬化的保护作用及其机制研究。方法:SD大鼠60只随机分为空白组、模型组、辛伐他汀组、龙眼叶总黄酮高剂量组、龙眼叶总黄酮低剂量组。空白组给予普通饲料喂养,其余动物采用高脂饲料喂养联合维生素D3腹腔注射方法建立动脉粥样硬化模型,连续饲养8周。各组动物给予相应药物灌胃,空白组与模型组给予同体积生理盐水,连续给药8周。末次给药2 h后,动物处死,取血清酶联免疫吸附法检测三酰甘油(triglyceride,TG)、总胆固醇(total cholesterol,TC)、低密度脂蛋白胆固醇(low density lipoprotein cholesterin,LDL-C)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化酶(glutathione peroxidase,GSH-Px)、丙二醛(malondialdehyde,MDA)、NO、氧化修饰低密度脂蛋白(oxidized low density lipoprotein,ox-LDL)、内皮素-1(endothelin-1,ET-1)、血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、C-反应蛋白(C-reactive protein,CRP)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)水平;取全血检测血液流变学各项指标。结果:与模型组比较,辛伐他汀组及龙眼叶总黄酮高剂量、低剂量均可不同程度降低模型动物血清中TC、TG、LDL-C、ox-LDL、ET-1、AngⅡ、TNF-α、IL-1β、CRP、IL-6、MDA水平,升高HDL-C、SOD、GSX-PX、NO水平,改善模型动物血液流变学相关指标,差异均有统计学意义(P<0.05或P<0.01)。结论:龙眼叶总黄酮对动脉粥样硬化模型大鼠具有一定的保护作用,可显著降低大鼠血脂水平、氧化应激反应、炎症因子水平,改善血液流变学相关指标。 Objective:To observe the protective effect of total flavonoids of longan leaves(TFLL)on atherosclerosis in rats and its mechanism.Methods:60 SD rats were randomly divided into blank group,model group,simvastatin group,total flavonoids of longan leaves high-dose group,and total flavonoids of longan leaves low-dose group.The blank group was fed with ordinary feed,and the rest of the animals were fed with high-fat feed combined with intraperitoneal injection of vitamin D3 to establish an atherosclerosis model,and kept for 8 weeks.The animals in each group were given the corresponding drugs by gavage.The blank group and the model group were given the same volume of normal saline for 8 weeks.Two hours after the last administration,the animals were sacrificed,and serum enzyme-linked immunosorbent assay was used to detect triglyceride(TG),total cholesterol(TC),and low density lipoprotein cholesterin(LDL-C),high density lipoprotein cholesterol(HDL-C),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA),NO,oxidized low density lipoprotein(ox-LDL),endothelin-1(ET-1),angiotensinⅡ(AngⅡ),C-reactive protein(CRP),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)levels;take whole blood to test hemorheology various indicators.Results:Compared with the model group,the simvastatin group and the total flavonoids of longan leaveshigh-dose group and low-dose group can reduce the TC,TG,LDL-C,ox-LDL,ET-1,AngⅡ,TNF-α,IL-1β,CRP,IL-6 and MDA levels,increased HDL-C,SOD,GSX-PX and NO levels,and improved hemorheology related indicators in model animals,and the differences were statistically significant(P<0.05 or P<0.01).Conclusion:The total flavonoids of longan leaves have a certain protective effect on atherosclerosis model rats,which can significantly reduce the levels of blood lipids,oxidative stress,and inflammatory factors in rats,and improve blood rheology related indicators.
