The Polymorphism Analysis between M229 and M307 Locus of FUT1 Gene in Three Group Pigs

摘要 为了探究3种显著差异类型猪FUT1基因M229与M307位点多态分布规律。本实验采用PCR-SSCP、PCR-RFLP结合直接测序技术分别对从江香猪、巴克夏×黔北黑猪F1代杂交猪和杜×长×大外三元猪群体M229和M307位点多态性进行分析。结果表明,从江香猪M229位点中有效等位基因为1.913 8,多态信息含量为0.363 5,属于中度多态,而在巴克夏×黔北黑猪F1代杂交猪和杜×长×大外三元猪群体中多态信息含量均低于0.25,为低度多态;与M229位点群体遗传参数结果相反,从江香猪M307位点多态信息含量最低,呈现出偏态分布,而在杜×长×大外三元猪群体中M307位点多态信息含量为0.260 2,可提供较多的遗传信息。因此,今后在从江香猪群体中应以M229位点选育为主,而国外品种应以M307位点选育为主。 In order to explore the polymorphic distribution of M229 and M307 locus of FUT1 gene in three significant group pigs. PCR-SSCP, PCR-RFLP combined with direct sequencing technology were used to observe the polymorphisms of M229 and M307 site in Congjiang pigs, the F1 hybrid pigs from the Berkshire × Qianbei black pig and the commercial crossbred pigs. The results showed that the effective allele was 1.913 8 from the M229 locus in Congjiang pigs, and the polymorphic information content was 0.363 5, which was moderate polymorphism, while the polymorphic information content of the F1 hybrid pigs from the Berkshire×Qianbei black pig and the commercial crossbred pigs was less than 0.25, which belong to low polymorphism. Contrary to the results of genetic parameters of M229 locus, the polymorphic information content of M307 locus from Congjiang pigs was the lowest, showing a skewed distribution, while the polymorphic information content of M307 locus in the commercial crossbred pigs was 0.260 2, which provides more genetic information. Therefore, in the future, the breeding of Congjiang pigs should be based on M229 locus, while foreign varieties should be selected mainly by M307 locus.
