响应面法优化超声提取鸡血藤总黄酮 被引量:3

On the Response Surface Method to Optimize Ultrasonic Extraction of Total Flavonoid from Caulis Spatholobi

摘要 鸡血藤为广西的道地药材,其主要活性成分为黄酮类物质。临床试验证明,鸡血藤中的黄酮类化合物具有抗肿瘤、抗病毒、降血脂等作用。该试验选用超声波辅助提取鸡血藤中总黄酮,以乙醇为溶剂,分别对超声时间、乙醇体积分数、料液比进行单因素分析;并在单因素试验的基础上,采用响应面法考察三者交互作用对总黄酮提取率的影响。利用软件Design-Expert8.0.6对试验数据进行回归分析,得到二次多项回归模型。结果表明:提取鸡血藤总黄酮提取的最佳工艺为超声时间41 min、料液比1∶52、乙醇体积分数41%,此条件下总黄酮的提取率为6.19%,与预测值基本一致。 Caulis Spatholobi is a native medicine of Guangxi.Its main active constituent is flavonoid.Clinical trials have proved that flavonoid compounds from Caulis Spatholobi have the functions of anti-tumor,antiviral,lipid,and so on.In this experiment,ultrasonic auxiliary is applied to extract the total yellow ketone from Caulis Spatholobi.Taking ethanol as solvent,a single factor analysis of ultrasound time,ethanol volume fractions and material liquid proportions is made seperatively.On the basis of single factors test,the response surface method is adopted to check the influence of the interaction of the three factors on the extraction rate of total yellow ketone.By using Design-Expert8.0.6,a regression analysis of the experimental data is made,and a quadratic regression model is obtained.The results show that the extraction technique of extracting total flavonoids from spatholobus suberectus is as the followings:41 minutes of ultrasonic time,1∶52 of material liquid proportion,41%of ethanol volume fraction,extraction of total flavonoids in this condition is 6.19%,consistent with the predicted values.In such a technique,the extraction rate of total flavonoids is 6.19%,which is nearly the same as the expected value.
