摘要
为研究水稻OsMPK7基因的功能,利用免疫印迹(Western blot,WB)技术检测水稻苗期OsMPK7蛋白质的表达特征,发现旱胁迫使OsMPK7蛋白质表达下调,提示OsMPK7基因在水稻旱胁迫应答中发挥作用。为了进一步验证其功能,构建了OsMPK7超表达载体,用农杆菌介导的方式转化水稻,通过PCR和WB鉴定,获得超表达纯合株系。拟旱胁迫条件下,超表达转基因水稻的种子萌发较晚,芽长与根长都显著低于野生型。苗期的缺水试验结果表明,在干旱胁迫下,超表达转基因水稻生长弱于野生型,出现明显的萎蔫现象。由此可见,超表达OsMPK7蛋白质降低了水稻对旱胁迫的耐受性。
In order to study the function of OsMPK7 gene in rice,the expression patterns of OsMPK7 protein was investigated by Western blot(WB).It was revealed that the expression abundance was decreased under drought stress,which suggested that the OsMPK7 protein played a role in response to drought stress.To further verify its function,a plasmid for over-expression OsMPK7 gene was constructed and transformed into rice variety TP309.Homozygous over-expression transgenic lines were identified by PCR and WB assay.Under drought stress,the seed germination took longer time in OsMPK7 over-expressed transgenic rice lines,and the shoot length and root length were significantly lower than those of wild type.The phenotypic observation showed that the growth of over-expressed transgenic rice was weaker than that of wild-type under drought stress.It can be concluded that the over-expression of OsMPK7 impaired the tolerance to drought stress.
