摘要
目的探讨长链非编码RNA NEAT(LncRNA NEAT1)及炎症因子白细胞介素(IL)-6和肿瘤坏死因子(TNF)-α在肺结核中的表达水平及诊断价值。方法根据《WS 288-2017肺结核诊断》标准,纳入活动性肺结核患者15例(活动性结核组),健康对照组22例,收集外周血,分离单个核细胞,利用实时荧光定量PCR检测外周血单个核细胞内LncRNA NEAT1、TNF-α、IL-6的表达水平,并采用Graphpad prism 5进行数据处理和绘制受试者工作特征(ROC)曲线。结果活动性结核组外周血单个核细胞中LncRNA NEAT1表达升高,且与健康对照组比较,差异有统计学意义(P<0.001)。细胞因子IL-6表达水平升高,与健康对照组比较,差异有统计学意义(P<0.05);TNF-α在活动性结核组表达水平上调,与健康对照组相比差异有统计学意义(P<0.01)。ROC曲线分析表明单项指标评价中,LncRNA NEAT1-1的特异度和灵敏度均为最高,分别为95.45%和93.33%,曲线下面积(AUC)达到0.9697。结论活动性肺结核患者LncRNA NEAT1、IL-6、TNF-α表达水平升高,LncRNA NEAT1可能通过调控炎症因子表达参与结核的发生、发展过程。
Objective To investigate the expression levels and diagnostic value of long-chain non-coding RNA NEAT(LncRNA NEAT1),inflammatory factors interleukin(IL)-6 and tumor necrosis factor(TNF)-αin tuberculosis.Methods According to the"WS 288-2017 Diagnosis of Tuberculosis"criteria,15 patients with active tuberculosis(active tuberculosis group)and 22 healthy controls(healthy control group)were enrolled.Peripheral blood was collected and mononuclear cells were isolated.Fluorescence quantitative PCR was used to detect the expression levels of LncRNA NEAT1,TNF-α,IL-6 in peripheral blood mononuclear cells,and Graphpad prism 5 was used for data processing and receiver operating characteristic(ROC)curve drawing.Results The expression of LncRNA NEAT1 in peripheral blood mononuclear cells of active tuberculosis group was increased when compared with healthy control group,and the difference was statistically significant(P<0.001).The expression level of cytokine IL-6 increased,compared with the healthy control group,and the difference was statistically significant(P<0.05).The expression level of TNF-αwas up-regulated in the active tuberculosis group,and the difference was statistically significant compared with the healthy control group(P<0.01).ROC curve analysis showed that in the evaluation of individual indicators,LncRNA NEAT1-1 had the best specificity and sensitivity,respectively 95.45%and 93.33%,and the area under the curve(AUC)was 0.9697.Conclusion The expression levels of LncRNA NEAT1,IL-6 and TNF-αare elevated in patients with active pulmonary tuberculosis.LncRNA NEAT1 may be involved in the occurrence and development of tuberculosis by regulating the expression of inflammatory factors.
