摘要
目的:探讨葛根对细菌性腹泻模型小鼠外周血浆Th1、Th2、Th17表达的影响,并观察其对胃肠传输功能的作用。方法:制备胃肠感染模型小鼠20只,随机分为模型组10只及干预组10只,干预组利用葛根水溶性生物碱灌胃,连续7 d;再将10只正常小鼠设为空白组。比较各组腹泻次数、外周血中Th1、Th2、Th17、血细胞表达水平,IL-2、IFN-γ、IL-4、IL-10、IL-17A、胃动素、血管活性肠肽的表达水平以及小鼠结肠传输时间。结果:造模后小鼠腹泻次数明显增加,造模后小鼠Th1、Th2细胞百分比、IL-2、IFN-γ、IL-4、IL-10、胃平滑肌层Obestatin及其受体GPR39、结肠传输时间明显下降,白细胞数量、中性粒细胞百分比、淋巴细胞百分比、Th17细胞百分比、IL-17A升高,与空白组比较差异均有统计学意义(P<0.05),经过葛根干预后上述指标明显改善,与模型组比较差异有统计学意义(P<0.05)。结论:葛根可明显改善胃肠感染,其作用机制与介导机体免疫反应有关。
Objective:To explore the effect of Pueraria lobata on the expression of Th1,Th2 and Th17 in peripheral plasma of mice with bacterial diarrhea,and to observe its effect on gastrointestinal transport function.Methods:Twenty mice with gastrointestinal infection were randomly divided into model group(n=10)and intervention group(n=10).The intervention group was fed with Pueraria water-soluble alkaloids for 7 days,and 10 normal mice were set as blank group.The times of diarrhea,the expression levels of Th1,Th2,Th17 and blood cells in peripheral blood,the expression levels of IL-2,IFN-γ,IL-4,IL-10,IL-17A,motilin,vasoactive intestinal peptide and colonic transit time in mice were compared.Results:After the establishment of the model,the number of diarrhea in mice increased significantly,and the percentage of Th1,Th2 cells,IL-2,IFN-γ,IL-4,IL-10,obestatin and its receptor GPR39 in gastric smooth muscle layer and colonic transit time decreased significantly,while the number of white blood cells,neutrophil percentage,lymphocyte percentage,Th17 cell percentage and IL-17A increased significantly compared with the blank group.After the intervention of Pueraria lobata,the above indexes were significantly improved compared with the model group(P<0.05).Conclusion:Pueraria lobata can significantly improve gastrointestinal infection,and its mechanism is related to mediating immune response.
