复方扶芳藤合剂对糖尿病模型大鼠BMSCs动员的影响 被引量:4

摘要 目的:探究复方扶芳藤合剂对糖尿病模型大鼠骨髓来源干细胞BMSCs动员的影响。方法:采用糖尿病模型大鼠作为实验模型,以随机数表法将糖尿病模型大鼠随机分为模型组、复方扶芳藤合剂组和西药组。模型组常规饲养;复方扶芳藤合剂组予复方扶芳藤合剂3.1 mL/d灌胃,一天灌胃两次,持续干预14天。西药组予集落刺激因子皮下注射,每两天注射一次,每次10μg/(kg×d),共计8次14天。以SD大鼠作为空白组。于干预14 d进行指标观察,流式细胞仪检测外周血中骨髓间充质干细胞(BMSCs)数量;ELISA检测外周血粒细胞集落刺激因子(G-CSF)含量变化。结果:流式细胞仪分析显示,与模型组相比较,复方扶芳藤合剂组外周血BMSCs数量明显加(P<0.05)。ELISA检测显示,与模型组相比较,复方扶芳藤合剂治疗14 d后,外周血G-CSF明显增加(P<0.05)。结论:复方扶芳藤合剂能发挥糖尿病模型大鼠骨髓间充质干细胞动员的作用,其机制可能与增加糖尿病大鼠外周血G-CSF含量有关。 Objective:To explore the effect of Fufangteng combination formula on BMSCs mobilization of bone marrow-derived stem cells in diabetic rats.Methods:Using the diabetes model rat as the experimental model,the diabetic rats were randomly divided into model group,compound Fufangteng combination formula group and western medicine group.The model group was given routine feeding;the Fufangteng combination formula group was gavage to the Fufangteng combination formula 3.1 mL/d,twice a day,and the intervention continued for 14 days.The western medicine group was injected with colony stimulating factor subcutaneously once every two days,10μg/(kgd)for 8 times for 14 days.SD rats were used as blank group.After 14 days of intervention,the number of bone marrow mesenchymal stem cells(BMSCs)in peripheral blood was detected by flow cytometry,changes of granulocyte colony stimulating factor(G-CSF)in peripheral blood were detected by ELISA.Results:The flow cytometer analysis showed that compared with the model group,the number of BMSCs in peripheral blood of the Fufangteng combination formula group was significantly increased(P<0.05).ELISA tests showed that compared with the model group,G-CSF in peripheral blood increased significantly after 14 days of treatment with Fufangteng combination formula(P<0.05).Conclusion:Fufangteng combination formula has a certain mobilization effect on BMSCs in diabetic rats,and its mechanism may be related to the increase of G-CSF content in peripheral blood of diabetic rats.
