桂南木莲和马关木莲正反交杂种的ISSR鉴定

Identification on Reciprocal Cross Offspring of Manglietia chingii and Manglietia maguanica by Using ISSR Markers

摘要 以桂南木莲、马关木莲及两者的杂交后代(正反交)共32株为试验材料,运用ISSR分子标记技术鉴定杂交后代的真实性。结果如下:从16条引物中筛选出4条引物用于杂种鉴定,试验表明30株杂交后代为真杂种,真实杂种率为100%;4条引物在正交杂种和亲本中扩增出46个位点,多态性位点率(PPB)为84.78%,材料间遗传相似系数为0.476~0.863,变幅为0.387。遗传相似系数0.714把亲本和正交种分为偏母本型、中间型和父本;4条引物在反交种和亲本中扩增出44个位点,多态性位点率(PPB)为88.78%,材料间遗传相似系数为0.400~0.872,变幅为0.472。遗传相似系数0.594把亲本和反交种分为偏母本型和偏父本型。30株杂交后代遗传多样性丰富,是培育木兰科新品种培育的重要材料。 The authenticity of the 32 Phybrid progenies of Manglietia chingii,Manglietia maguanica and their hybrids(inverse and obverse cross)was identified by ISSR molecular markers.The results showed that 30 hybrids were true hybrids,and the rate of true hybridization was 100%.Four primers were screened from 16 primers for hybrid identification.A total of 46 loci were amplified by four primers in obverse hybrids and parents,with the percentage of polymorphic loci(PPB)of 84.78%.The genetic similarity coefficient among materials ranged from 0.476 to 0.863,with the variation range was 0.387.The genetic similarity coefficient was 0.714,dividing the parents and obverse hybrids into maternal type,intermediate type and paternal type.A total of 44 loci were amplified by four primers in inverse hybrids and parents,in which 39 loci were polymorphic,with the percentage of polymorphic loci(PPB)of 88.78%.The genetic similarity coefficient among materials ranged from 0.400 to 0.872 and the variation range was 0.472.The genetic similarity coefficient 0.594,dividing the parents and inverse hybrids into maternal type and paternal type.The genetic diversity of thirty hybrid progenies are abundant.It is an important material for breeding new varieties of Magnoliaceae.
