摘要
目的:探讨白芷总香豆素治疗原发性痛经(PDM)的疗效,并揭示其可能的治疗机制。方法:采用缩宫素诱导的原发性小鼠痛经模型,以注射催产素后30 min内扭体次数和扭体潜伏期为主要观测指标,考察白芷总香豆素的镇痛作用。使用BL-420F生物机能实验系统观察白芷总香豆素对原发性痛经模型小鼠子宫收缩频率和幅度的影响及对缩宫素所致正常小鼠子宫平滑肌痉挛的对抗作用。测定血清中谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量,ELISA法检测血浆β-内啡肽,子宫组织前列腺素F 2α(PGF 2α)、6-酮-前列腺素F 1α(6-Keto-PGF 1α)和血栓素B 2(TXB 2)的含量。结果:与模型组相比,白芷总香豆素可通过降低PDM模型小鼠的扭体反应并呈剂量依赖性地显示镇痛活性,同时可明显减少原发性痛经模型小鼠离体子宫平滑肌收缩频率,降低收缩幅度。白芷总香豆素对缩宫素诱发正常小鼠子宫平滑肌痉挛性收缩有明显的抑制作用。白芷总香豆素也能显著降低子宫组织中PGF 2α的含量,降低血清中MDA的含量,升高子宫组织中6-keto-PGF 1α/TXB 2水平,提高血清GSH-Px,SOD的活力及血浆β-内啡肽含量。结论:白芷总香豆素能有效改善实验性PDM,其机制可能与抗氧化、降低PGF 2α水平,升高6-keto-PGF 1α/TXB 2水平及β-内啡肽含量有关。
Objective:To study the therapeutic effect of total coumarin of Angelica dahurica on primary dysmenorrhea(PDM)and to reveal its possible therapeutic mechanism.Methods:The PDM model was induced by oxytocin.The analgesic effect of total coumarin of Angelica dahurica was studied by observing the number of writhing and the latency of writhing within 30 minutes after oxytocin injection.The effects of total coumarin of Angelica dahurica on the frequency and amplitude of uterine contraction in mice with PMD and on the antispasmodic effect of uterine smooth muscle in normal mice induced by oxytocin were observed by BL-420F biological function system.The serum contents of GSH-Px,MDA and SOD were measured by kits.The plasma contents ofβ-endorphin,as well as PGF 2α,6-Keto-PGF 1αand TXB 2 in uterine tissue were measured by ELISA method.Results:Compared to the model group,the total coumarin of Angelica dahurica showed the analgesic activity in a dose-dependent manner by reducing the writhing response of the PDM model mice;meanwhile,it could significantly reduce the contractile frequency and amplitude of isolated uterine smooth muscle of PMD model mice.The total coumarin of Angelica dahurica also could obviously inhibit the spasmodic contraction of uterine smooth muscle induced by oxytocin in normal mice.It could significantly reduce the content of PGF 2αin uterine tissues,reduce the content of MDA in serum,increase the level of 6-keto-PGF 1α/TXB 2,and increase the activity of GSH-Px and SOD in serum and the content ofβ-endorphin in plasma.Conclusion:The total coumarin of Angelica dahurica can effectively improve the experimental PDM,and its mechanism may be related to antioxidation,decrease of PGF 2αand increase of 6-keto-PGF 1α/TXB 2 level andβ-endorphin content.
