摘要
目的:研究桂参通脉胶囊对心衰大鼠血管紧张素Ⅱ(AngⅡ)和醛固酮(ALD)的影响,探讨其在防治心力衰竭中的作用机制。方法:采用腹腔注射阿霉素的方法建立心力衰竭大鼠模型,随机将大鼠分为心衰模型组、桂参通脉胶囊高、中、低剂量组、卡托普利组及空白对照组,检测各组心衰大鼠血浆AngⅡ、ALD水平及血清NT-proBNP浓度差异,并观察心脏组织形态变化。结果:各组血浆AngⅡ、ALD及血清NT-proBNP浓度皆高于空白对照组,桂参通脉胶囊高、中剂量组和卡托普利组血浆AngⅡ、ALD升高较少,桂参通脉胶囊高剂量组血清NT-proBNP浓度最低;同时观测到各组均有不同程度心肌细胞凋亡,桂参通脉胶囊中、高剂量组和卡托普利组心肌细胞损伤较心衰模型组有明显改善,其光镜下病理评分差异有统计学意义(P<0.05)。结论:桂参通脉胶囊能够降低血浆AngⅡ、ALD的水平,抑制神经内分泌系统的过度激活,延缓心室重构;降低血清NT-proBNP,改善心功能;减少心肌细胞病理改变,减缓心肌细胞凋亡。
Objective:To study the effects of Guishen Tongmai Capsule on angiotensinⅡ(AngⅡ)and aldosterone(ALD)in rats with heart failure,and to explore its action mechanism in the prevention and treatment of heart failure,thus to effectively develop the new drug of Guishen Tongmai Capsule.Methods:Rat model of heart failure was established by intraperitoneal injection of Adriamycin solution.The rats were randomly divided into the model group,Guishen Tongmai Capsule groups of high-dose,medium-dose and low-dose,Captopril group and the blank control group.The plasma levels of AngⅡand ALD,as well as the serum concentration of NT-proBNP were measured in each group after the interventions.The changes of cardiac tissue morphology were observed as well.Results:The plasma levels of AngⅡand ALD,as well as the serum concentration of NT-proBNP were higher after modeling compared to those in the blank control group.After interventions,the plasma levels of AngⅡand ALD were lower in Guishen Tongmai Capsule groups of high-dose and medium-dose and in Captopril group;the serum concentration of NT-proBNP was the lowest in the high-dose Guishen Tongmai Capsule group.The apoptosis of cardiomyocytes was observed in each modeling group.After interventions,the myocardial cell damages in Guishen Tongmai Capsules of medium-dose and high-dose and in Captopril group were significantly improved than those in the model group(P<0.05).Conclusion:Guishen Tongmai Capsule can reduce the plasma levels of AngⅡand ALD,inhibit the excessive activation of neuroendocrine system and delay the ventricular remodeling.It can reduce serum NT-proBNP,improve cardiac function,reduce the pathological changes of cardiomyocytes and slow down the apoptosis of cardiomyocytes.
