摘要
为辣椒番茄斑萎病毒病绿色防控及分子育种提供理论依据,以贵州主栽辣椒品种百宜辣椒和遵义朝天椒及易感病品种茄门甜椒作为研究材料,采用ELISA法测定感染番茄斑萎病毒(TSWV)辣椒超氧化物岐化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、苯丙氨酸解氨酶(PAL)和多酚氧化酶(PPO)活性的变化,明确辣椒对TSWV的抗性与防御酶活性的关系。结果表明:接种TSWV第1天时,3个辣椒品种的PAL、POD、PPO和CAT活性均增高,PAL和CAT的最高峰出现在第7天,POD和PPO出现在第5天;SOD活性呈先升后降趋势,7 d后与对照(健康)的基本一致。遵义朝天椒和百宜辣椒PAL、POD、PPO和CAT活性出现2个峰值,茄门甜椒出现1个峰值,遵义朝天椒和百宜辣椒对TSWV的抗性较好,茄门甜椒的抗性稍弱。5种防御酶活性与辣椒抗TSWV密切相关,在辣椒对TSWV抗性生化机制中起重要作用。
To provide theoretical basis for green prevention and control of pepper tomato spotted wilt virus(TSWV)disease and molecular breeding,the main planting cultivars Baiyi pepper and Zunyi Chaotian pepper,and susceptible cultivar Qiemen pepper,were inoculated with TSWV and tested through ELISA for the activity changes of superoxide disproportionation enzyme(SOD),peroxidase(POD),catalase(CAT)and phenylalanine ammonia enzyme(PAL)and polyphenol oxidase(PPO).Results:After inoculation,the activities of PAL,POD,PPO and CAT of the three pepper varieties all increased at 1 st day,the peak of PAL and CAT appeared on the 7 th day,and the peak of POD and PPO appeared on the 5 th day.SOD activity increased first and then was basically the same as the control activity.The activities of PAL,POD,PPO and CAT of Zunyi Chaotian pepper and Baiyi pepper showed two peaks,and the activity of Qiemen pepper showed one peak,indicating that Zunyi Chaotian pepper and Baiyi pepper had better resistance to TSWV,while Qiemen pepper had relatively lower resistance to TSWV.The activities of the five defendant enzymes,PAL,POD,PPO,CAT and SOD,were closely related to the resistance to TSWV,and played an important role in the biochemical mechanism of the resistance to TSWV of pepper.
