摘要
采用不同植物生长调节剂组合对绿洲一号菌草主茎或侧枝上的幼茎(宝塔状)侧芽和茎尖进行初代芽分化诱导、继代增殖和生根培养,以建立绿洲一号菌草的快繁技术体系。结果表明,幼嫩茎段侧芽在MS+3.0 mg/L6-BA+0.2 mg/L 2,4-D+1.0 mg/L IBA培养基中进行初代培养,可平均分化2.64个幼芽(苗),分化的丛生苗和分蘖苗健壮均匀;最佳继代增殖培养基为MS+5.0 mg/L 6-BA+1.0 mg/L IBA,在此培养基上继代增殖,增殖系数达到6.12,增殖的幼芽密集、健壮,生长势强;最佳生根培养基为MS+5.0 mg/L IBA,小芽苗在此培养基上诱导生根,平均根数为11.75条,根长为3.33 cm,根细而密,有韧性。
By studying the effects of different combinations of plant growth regulators on the induction of primary bud differentiation,subgenerational proliferation and rooting culture of the lateral buds and steam tips from main stem or lateral brunch's young stem(pagoda-shaped)of Lüzhou 1 fungus grass,the technique system of tissue culture and rapid propagation of Lüzhou 1 fungus grass was established.The results showed that when the lateral buds of the young stem segment were cultured in MS+3.0 mg/L 6-BA+0.2 mg/L 2,4-D+1.0 mg/L IBA medium,the average number of shoots was 2.64,and the clustered seedlings and tiller seedlings were robust and uniform.The optimal proliferation medium was MS+5.0 mg/L 6-BA+1.0 mg/L IBA with proliferation coefficient of 6.12,and the proliferative buds were dense,robust and had strong growth potential.The optimum rooting medium was MS+5.0 mg/L IBA,under this medium,the average number of roots was 11.75,and the length of roots was 3.33 cm,meanwhile the roots were thin,dense and tough.
