摘要
目的研究海菖蒲黄酮FEA(Enhalus acoroides flavonoids,FEA)对LPS刺激RAW264.7细胞引起的炎症反应的改善作用。方法用1µg/mL的LPS刺激RAW264.7细胞6 h后,用含有不同浓度的海菖蒲黄酮FEA作用RAW264.7细胞20 h,采用ELISA方法测定RAW264.7细胞分泌的NO、TNF-α、IL-1β和IL-6的含量,采用western-blot方法检测NF-ĸB信号通路中IĸBα、P-IĸBα、p65、P-p65的蛋白表达含量。结果海菖蒲黄酮FEA可以抑制由LPS刺激RAW264.7细胞引起的NO、TNF-α、IL-1β和IL-6细胞因子的分泌,减少磷酸化蛋白IĸBα和p65的表达量。结论海菖蒲黄酮FEA可以减少细胞炎症因子的产生和抑制NF-ĸB信号通路的活化来改善炎症作用。
Objective To study the anti-inflammatory activity of flavonoids from Enhalus acoroides induced by LPS on RAW264.7 cells.Methods After being stimulated with 1μg/mL LPS for 6 h,RAW264.7 cells were further treated with different concentrations of flavonoids for additional 20 h.The amount of secreted NO,TNF-α,IL-1βand IL-6 were measured by ELISA method.The expression level of IκBα,P-IκBα,p65 and P-p65 were measured using western-blot method.Results The Enhalus acoroides flavonoids could inhibit the secretion of cytokines of NO,TNF-α,IL-1βand IL-6,and reduce the expression of phosphorylated proteins of IκBαand p65.Conclusion The Enhalus acoroides flavonoids could exert anti-inflammatory effects by reducing the expression of cytokines and inhibiting activation of NF-κB signaling pathway.
