摘要
目的利用响应面法优化多序岩黄芪多酚的提取工艺。方法以多序岩黄芪药材粉末为原料,在单因素实验基础上,利用超声波辅助提取,考察乙醇浓度、料液比、提取功率和提取时间对多序岩黄芪中多酚提取率的影响,根据Box-Behnken实验结果,采用响应面法对多序岩黄芪多酚类工艺进行优化。结果确定最佳提取工艺参数:提取溶剂为60%乙醇、料液比为1:29(g:mL)、提取温度为55℃、超声功率为120 W和超声时间为40min。此提取条件下多序岩黄芪多酚的含量达5.127mg/g。结论该工艺具有简单可行,重复性好的优点,适用于多序岩黄芪多酚的提取。
Objective To optimize the extraction process of polyphends from Hedysarum polybotrys Hand. by response surface methodology. Methods Taking Hedysarum polybotrys Hand. roots as raw material, the effects of ethanol concentration, solid-liquid ratio, extraction power and extraction time on the extraction yield of polyphenols from Hedysarum polybotrys Hand. were investigated by ultrasonic assisted extraction on the basis of single factor test. According to the results of Box-Behnken experiment, the response surface method was used to optimize the polyphenolic process of Hedysarum polybotrys Hand. Results The optimum extraction process parameters were showed as follows: the ethanol concentration was 60%, solid-liquid ratio was 1:29(g:mL), extraction temperature was 55 °C, ultrasound power was 120 W and ultrasound time was 40 min, and the polyphenol content was determined as 5.127 mg/g. Conclusion This process has the advantages of simplicity, feasibility, and good repeatability, and is suitable for the extraction of polyphenols of Hedysarum polybotrys Hand.
