摘要
目的观察枸杞多糖对糖尿病肾病大鼠肾功能的保护作用,探讨TLR4/Myd88信号通路的作用机制。方法 SD大鼠45只,随机分为假手术组(Sham组),糖尿病肾病模型组(DN组)、枸杞多糖治疗组(LBP组),DN组和LBP组采用三联法(手术+高糖高脂+STZ)法建立Ⅱ型糖尿病肾病大鼠模型,建模后LBP组给予枸杞多糖100mg/Kg灌胃,Sham组及HS组予以等量生理盐水;血生化检测大鼠肌酐(Scr)、血尿素氮(BUN)浓度,ELISA检测炎性因子TNF-α、IL-6、IL-10的水平,HE染色观察肾脏组织病理学变化,qPCR、Western blot检测肾脏组织中TLR4、Myd88、NF-κB表达。结果糖尿病肾病大鼠可导致肾功能损伤,肾小球细胞结构破坏,炎性因子侵润;枸杞多糖可以减轻糖尿病肾病大鼠的肾损伤,降低炎症反应,改善肾功能,且可下调TLR4、Myd88、NF-κB蛋白表达。结论枸杞多糖减轻糖尿病大鼠肾功能损伤,与TLR4/Myd88信号通路介导炎症反应下降有关。
bjective To study the protective effect of Lycium barbarum polysaccharides on renal function in rats with diabetic nephropathy,and to explore the relationship with TLR4/Myd88 signaling pathway.Methods Forty-five SD rats were randomly divided into sham operation group(Sham group),diabetic nephropathy model group(DN group),Lycium barbarum polysaccharide treatment group(LBP group),rats in DN group and LBP group were treated by triple method(surgery+high glucose high fat).The rat model of type-Ⅱdiabetic nephropathy was established by lipid+STZ method.LBP group was given 100 mg/Kg of sputum polysaccharide,Sham group and HS group were given normal saline.The levels of Scr and BUN were detected by blood biochemical examination,ELISA was used to detect the inflammatory factors TNF-α,IL-6,IL-10,HE staining was used to observe the renal histopathological changes,QPCR and Western blot were used to examine the TLR4,Myd88,NF-κB expression.Results The renal dysfunction,glomerular cell structure destruction and inflammatory factor infiltration were found in diabetic nephropathy rats.Lycium barbarum polysaccharide alleviated kidney damage,reduced inflammation response and improved renal function in rats with diabetic nephropathy,downregulated the expression levels of TLR4,Myd88 and NF-κB proteins.Conclusion Lycium barbarum polysaccharide alleviated renal function damage is related to reduction of inflammatory response via TLR4/Myd88 signaling pathway in diabetic rats..
