摘要
目的研究中药藤梨根提取物的体外抗肿瘤转移作用。方法藤梨根药材采用95%乙醇溶剂回流提取,通过MTT实验、划痕实验、Transwell实验和明胶酶谱法评价不同浓度藤梨根95%乙醇提取物对人乳腺癌细胞(MDA-MB-231)的抗肿瘤转移作用。结果体外抗肿瘤转移活性筛选实验结果表明,藤梨根95%乙醇提取物在100μg/mL浓度以下对MDA-MB-231细胞没有细胞毒性,在不同浓度下能明显抑制MDA-MB-231细胞的迁移和侵袭,并呈一定剂量依赖性,IC50值分别为14.222、16.973μg/mL。明胶酶谱法实验结果表明,藤梨根95%乙醇提取物抗肿瘤转移作用与抑制基质金属蛋白酶MMP-9活性有关。结论藤梨根提取物具有较强的体外抗肿瘤转移活性。
Objective To study the anti-tumor metastasis effect in vitro of extracts from actinidia arguta,which is a traditional Chinese medicine.Methods The actinidia arguta was extracted with 95%ethanol solvent refluxing process.MTT assay,scratch test,Trans well assay and gelatin zymography method were used to evaluate the anti-tumor metastasis effect of different concentrations of 95%ethanol extracts from actinidia arguta on human breast cancer cells(MDA-MB-231).Results The results of in vitro anti-tumor metastasis activity screening experiments showed that the 95%ethanol extracts from actinidia arguta had no cytotoxicity to MDA-MB-231 cells below the concentration of 100μg/mL,and under different concentrations it could significantly inhibit the migration and invasion of MDA-MB-231 cells in a dose-dependent manner,with IC50 values of 14.222μg/mL and 16.973μg/mL respectively.The results of gelatin zymography method assay showed that the anti-tumor metastasis effect of 95%ethanol extracts from actinidia arguta was related to the inhibition of matrix metalloproteinase-9 activity.Conclusion Extracts from actinidia arguta have strong anti-tumor metastasis activities in vitro.
