摘要
骨再生修复是临床医学包括口腔医学面临的重大难题。近年来,越来越多的研究表明二甲双胍(Metformin,Met)具有促成骨作用,有望应用于牙周组织再生领域。然而,其促成骨的机制迄今仍未完全清楚。目前认为,二甲双胍主要通过激活单磷酸腺苷活化蛋白激酶(Adenosine 5’-monophosphate activated protein kinase,AMPK)通路而发挥促成骨作用;此外,二甲双胍还可磷酸化细胞外信号调节激酶(extracellular signal-regulated Kinase,ERK),刺激内皮和诱导型一氧化氮合酶(endothelial and inducible nitric oxide synthases,e/iNOS),抑制GSK3β/Wnt/β-catenin通路,并促进成骨细胞的成骨分化。本文将主要对二甲双胍的促成骨作用进行阐述。
Bone regeneration and repair is a major problem in clinical medicine,including Stomatology.A number of studies in recent years have shown that metformin has effect of promotingosteogensis,suggesting that metformin may be used in periodontal tissue regeneration.However,the mechanism of its osteogenic promoting effect has not been fully elucidated.At present,metformin plays an important role in promoting osteogensis by activating the adenosine monophosphate activated protein kinase(AMPK)pathway.In addition,Metformin induced activation of phosphorylated extracellular signal-regulated kinase(ERK)and stimulated the expression of endothelial and inducible nitric oxide synthases(e/iNOS),which also can promote osteogensis.These mechanisms will be discussed in detail in this review.
