摘要
目的:探究针刺慢性温和刺激(CUMS)大鼠过程中粪便关键代谢通路。方法:刺激大鼠24只,空白大鼠6只。刺激大鼠进行CUMS,4周后将刺激大鼠均分为4组:CUMS组、针刺组(CUMS+1W组、CUMS+2W组和CUMS+3W组)。开始针刺,1周后CUMS+1W组取材,2周后CUMS+2W组取材,3周后CUMS+3W组和CUMS组取材。取盲肠内容物检测(GC-MS)。结果:造模过程中刺激大鼠糖水消耗数值不断下降,4周后显著低于空白大鼠。针刺过程中,针刺大鼠糖水消耗呈现上升趋势,直到针刺3周后,针刺大鼠与空白大鼠差异无统计学意义。分析粪便差异物,38种在CUMS组和CUMS+3W组中呈现差异有统计学意义,其中23种代谢物在治疗过程中具有一致的变化趋势(升高或降低)。主要与以下途径相关:甘、苏和丝氨酸的代谢,氨酰基的合成,蛋氨酸和半胱氨酸的代谢和磷酸肌醇代谢。结论:以上可能是针刺的粪便关键代谢通路。
Objective:To explore the key fecal metabolic pathways of acupuncture in treating CUMS in rats.Methods:Chronic unpredictable mild stimulation(CUMS)model was established in 24 rats,another 6 rats were taken as the blank control.After four weeks of modeling,the stimulated rats were randomly divided into the model group,the acupuncture groups of one week,two weeks and three weeks.The samples were collected after one week of acupuncture,two weeks of acupuncture and three weeks of acupuncture.GC-MS analysis was used to detect the contents of the cecum in rats.Results:Sucrose preference test of the stimulated rats decreased continuously during the modeling process,and were significantly lower than that of the blank control rats after four weeks.During the acupuncture,the sucrose preference test of the acupuncture rats showed an upward trend.After three weeks of acupuncture,there was no statistical difference between the acupuncture group and the control group.Analysis was performed on feces,38 were significantly different between the model group and the acupuncture group of three weeks;of these 38 differential metabolites,23 metabolites had a consistent increasing or decreasing change trend during the treatment process.These metabolites pathways related involved the metabolisms of glycine,serine and threonine,the biosynthesis of aminoacyl,the metabolisms of methionine,cysteine and phosphoinositide.Conclusion:Above all may be the key metabolic pathways of feces when treated by acupuncture.
