电针联合身痛逐瘀胶囊改善慢性坐骨神经损伤模型大鼠疼痛及对GFAP/p38MAPK信号通路的影响 被引量:2

Effect of Electro-Acupuncture Combined with Shentong Zhuyu Capsule on Pain in CCI Model Rats and Its Influence to GFAP/p38MAPK Signaling Pathway

摘要 目的:观察电针联合身痛逐瘀胶囊对慢性坐骨神经损伤(CCI)大鼠疼痛及脊髓组织胶质纤维酸性蛋白(GFAP)及磷酸化p38丝裂原活化蛋白激酶(p-p38MAPK)蛋白表达的影响。方法:90只清洁级SD大鼠随机分为假手术组(Sham组)、模型组(Model组)、电针组、中药组和针药组,每组分为3 d、7 d和14 d共3个亚组,每个亚组6只大鼠。按照Bennett的方法建立CCI大鼠模型,电针组给予损伤侧环跳、阳陵泉电针治疗,每日1次,每次30 min;中药组给予身痛逐瘀胶囊治疗(1.64 g/kg),药物通过灌胃给药,按照15 mL/kg的给药容积,每只大鼠每天灌胃1次;针药组在给予每日1次电针治疗的基础上,同时给予中药灌胃治疗;假手术组和模型组给予等容积生理盐水灌胃。采用热缩足反射潜伏期(TWL)检测大鼠行为学变化,Western blot法检测大鼠脊髓组织GFAP、p-p38MAPK蛋白表达。结果:TWL检测结果显示,模型组大鼠各时间点TWL值较假手术组显著降低(P<0.05);电针组、中药组及针药组各时间点TWL较模型组显著升高(P<0.05);针药组大鼠各时间点TWL较电针组和中药组显著较高(P<0.05)。Western blot检测结果表明,模型组大鼠各时间点GFAP、p-p38MAPK蛋白表达较假手术组显著降低(P<0.05);电针组、中药组及针药组各时间点GFAP、p-p38MAPK蛋白表达较模型组显著升高(P<0.05);针药组大鼠各时间点GFAP、p-p38MAPK蛋白表达较电针组和中药组显著较高(P<0.05)。结论:电针联合身痛逐瘀胶囊能够减轻CCI模型大鼠疼痛反应,其机制可能与下调脊髓组织中GFAP和p-p38MAPK蛋白表达有关。 Objective:To observe the effects of electro-acupuncture combined with Shentong Zhuyu Capsule on pain,glial fibrillary acidic protein(GFAP)of spinal cord and phosphorylated p38 mitogen-activated protein kinase(p-p38MAPK)protein in CCI rats.Methods:90 SD rats were randomly divided into the sham operation group,the model group,the electro-acupuncture group,the Chinese medication group and the combination group(electro-acupuncture+Chinese medication).Each group was subdivided into three groups according to 3d,7d and 14d three different time points,with 6 rats in each subgroup.The CCI rat model was established according to Bennett's method.The electro-acupuncture group was treated with electro-needling Huantiao(BL30)and Yanglingquan(GB34)on injury side,once a day for 30 min.The Chinese medication group was treated with Shentong Zhuyu Capsule(1.64 g/kg),each rat was intragastrically administered at the volume of 15 ml/kg once a day.The combination group was given the treatments of the electro-acupuncture group and the Chinese medication group.The sham operation group and the model group were given an equal volume of normal saline.The behavioral changes of rats were detected by TWL.The expressions of GFAP and p-p38MAPK protein in rat spinal cord tissue were detected by Western blot method.Results:The TWL test results showed that the TWL values of the rats in the model group were significantly lower than those in the sham operation group at each time point(P<0.05).The TWL values were significantly increased in the electro-acupuncture group,the Chinese medication group and the combination group compared to those in the model group at each time point(P<0.05);of which the increases were more significant in the combination group(P<0.05).Western blot analysis showed that the expressions of GFAP and p-p38MAPK protein in the model group were significantly lower than those in the sham operation group(P<0.05).The expressions of GFAP and p-p38MAPK protein were significantly increased in the electro-acupuncture group,the Chinese medication group and the combination group compared to those in the model group(P<0.05);of which the increases were more significant in the combination group(P<0.05).Conclusion:Electro-acupuncture combined with Shentong Zhuyu Capsule can alleviate the pain response in CCI model rats,and its mechanism may be related to down-regulation of GFAP and p-p38MAPK protein in spinal cord tissue.
