摘要
随着人们生活方式和环境因素的变化,癌症的发病率呈不断上升之势。二茂铁类化合物具有廉价、稳定、亲脂、强氧化还原和低毒等特性,显示出良好的抗肿瘤活性。其中,二茂铁-酚杂合体Ferrocifen对包括耐药肿瘤细胞系在内的多种肿瘤细胞系具有显著地抑制作用。当前,此类化合物正处于临床前研究阶段,有希望在不久的将来为人类健康服务。显然,二茂铁杂合体在抗肿瘤领域有着广泛的应用前景。本文将对近年来二茂铁杂合体在抗肿瘤领域中的研究进展作出概述,以求为药物化学家进一步设计活性更高的杂合体提供参考。
The morbidity of cancer continues to grow due to the change of lifestyle and environment.Ferrocene derivatives which endow with many advantages such as low cost,stability,high lipotropy,strong redox,and low toxicity,exhibit potential anticancer activity.Among them,ferrocene-phenol hybrid Ferrocifen which possess excellent activity against various cancer cell lines including drug-resistant cells,is in pre-clinical trial,demonstrating the anticancer potential of ferrocene hybrids.This review will summarize the recent development of ferrocene hybrids as potential anticancer leads to pave the way for the further design of novel hybrids with higher activity.
