3种miRNA在多囊卵巢综合征痰湿证血清中的表达及临床意义 被引量:5

Expression and Diagnostic Value of Three Circulating miRNAs in Polycystic Ovary Syndrome Patients with Phlegm-Dampness Syndrome

摘要 目的分析多囊卵巢综合征痰湿证患者血清中miR-642a、miR-2861、miR-146a的表达差异及生殖内分泌、糖脂代谢特征,探讨多囊卵巢综合征痰湿证产生机制。方法选取行体外受精-胚胎移植的不孕患者共63例,其中多囊卵巢综合征痰湿证21例(痰湿组)、多囊卵巢综合征非痰湿证19例(非痰湿组)、男方因素不孕症23例(对照组)。采用实时荧光定量PCR检测患者血清中3种miRNA的表达差异,分析比较3组患者临床特征、实验室指标和血清miRNA相对表达量的差异。结果①痰湿组BMI、WHR水平高于非痰湿组(P<0.05)。②痰湿组T、FINS、HOMA-IR、TC、TG、HDL、LDL水平较对照组具有显著性差异(P<0.05);非痰湿组LH水平较对照组显著升高(P<0.05);与非痰湿组比较,痰湿组FINS、HOMA-IR、TG、HDL-C水平具有显著性差异(P<0.05)。③3组间血清3种miRNA表达水平具有显著性差异(P<0.05),进一步两两比较显示PCOS较对照组、PCOS痰湿组较非痰湿组及对照组相对表达量均具有显著性差异(P<0.05)。结论miR-642a、miR-2861、miR-146a在多囊卵巢综合征患者,尤其是痰湿证患者血清中的特异性差异表达,可能参与了多囊卵巢综合征痰湿证的发病过程。 Objective To study the pathogenesis of phlegm-dampness syndrome in polycystic ovary syndrome by researchingthe expressions of miR-642 a,miR-2861 and miR-146 a in the serum,the characteristics of reproductive endocrine and the glycolipid metabolism of polycystic ovary syndrome(PCOS)patients.Methods A total of 63 infertile patients undergoing in vitro fertilization-embryo transfer were enrolled,including 21 PCOS patients with phlegm-dampness syndrome(phlegm-dampness group),19 patients with polycystic ovary syndrome without phlegm-dampness syndrome(non-phlegm-dampness group)and 23 infertile patients with male factor(control group).The expressions of three miRNAs in serum were detected by real-time fluorescentquantitative PCR.The clinical data,laboratory parameters and the relative expressions of serum miRNAs were compared among the three groups.Results①The levels of BMI and WHR in the phlegm-dampness group were higher than those in the non-phlegm-dampness group(P<0.05).②The levels of T,FINS,HOMA-IR,TC,TG,HDL and LDL in the phlegm-dampness group were significantly higher than those in the control group(P<0.05).The level of LH in non-phlegm-dampness group was significantly higher than that in the control group(P<0.05).Compared with non-phlegm-dampness group,the levels of FINS,HOMA-IR,TG and HDL-C in the phlegm-dampness group were significantly higher(P<0.05).③There were significant differences in the expression levels of three miRNAs among the three groups(P<0.05).The relative expression in PCOS patients,especially in the PCOS phlegm-dampness group was quite different from that of the control group and the non-dampness group(P<0.05).Conclusion The specific expressions of miR-642 a,miR-2861 and miR-146 a in the serum of patients with polycystic ovary syndrome,especially those with phlegm-dampness syndrome,may be involved in the pathogenesis of phlegm-dampness syndrome in polycystic ovary syndrome.
