摘要
目的探讨电针对功能性胃肠病(FGIDs)模型大鼠内脏敏感性的影响机制,及其脏腑穴位特异性。方法40只雄性SD大鼠随机分成正常组、模型组、足三里组、阴陵泉组、阴谷组,每组8只。采用综合方法复制FGIDs大鼠模型,电针足三里组、阴陵泉组、阴谷组,每日1次,1次30 min,连续针刺7 d,模型组只捆绑不针刺。电针后对大鼠行结直肠扩张刺激实验,进行腹部撤回反射(AWR)评分;后取大鼠下丘脑、脊髓(T5~10),用免疫印迹(Western blot)方法测定丝裂原活化蛋白激酶(p38 MAPK)的蛋白表达。结果模型组大鼠AWR评分较正常组升高(P<0.01);与模型组比较,在各压力刺激下足三里组评分均明显降低(P<0.05),阴陵泉组在20、60、80 mm Hg评分明显降低(P<0.05);与阴谷组比较,足三里组在各个压力刺激下评分均明显下降(P<0.05),阴陵泉组在80 mm Hg压力下差异有统计学意义(P<0.05);足三里组与阴陵泉组评分比较无显著性差异(P>0.05)。模型组p38蛋白活性表达较正常组明显升高(P<0.01);与模型组比较,足三里组、阴陵泉组p38蛋白活性表达明显降低(P<0.05);在下丘脑中:与阴谷组比较,足三里组p38降低明显(P<0.05);在脊髓中:与阴陵泉组、阴谷组比较,足三里组降低明显(P<0.05)。结论电针足三里穴和阴陵泉穴均可改善FGIDs模型大鼠AWR评分,降低其内脏敏感性,其作用机制可能与电针能够抑制p38 MAPK蛋白活化水平有关,且电针足三里穴对p38 MAPK表达的调节作用较阴陵泉穴更为明显,具有穴位特异性。
Objective To investigate the mechanism of electroacupuncture on visceral sensitivity of functional gastroenterology(FGIDs)model in rats,and the point specificity of viscera.Methods 40 male SD rats were randomly divided into normal group,model group,Zusanli group,Yinglingquan group and Yingu group,with 8 rats in each group.The FGIDs rat model was established by comprehensive method.Electroacupuncture was performed in Zusanli group,Yinglingquan group and Yingu group once a day for 30 min for 7 days.The model group only binding without acupuncture.After electroacupuncture,the rats were subjected to Colorectal dilatation test,and the abdominal withdrawal reflex(AWR)score was obtained.After that,the hypothalamus and spinal cord(T5-10)of the rats were taken,and the protein expression of mitogen-activated protein kinase(p38 MAPK)was determined by Western blot.Results The AWR score of rats in the model group was significantly increased compared with that in the normal group(P<0.01).Compared with the model group,the scores of Zusanli group were significantly reduced under various pressure stimuli(P<0.05),and those of Yinglingquan group were significantly reduced in 20,60 and 80 mm Hg(P<0.05).Compared with Yingu group,the scores of the Zusanli group were significantly decreased under various pressure stimuli(P<0.05),and the differences of the Yinlingquan group under 80 mm Hg pressure were statistically significant(P<0.05).There was no significant difference between Zusanli group and Yinglingquan group(P>0.05).The activity expression of p38 MAPK in the model group was significantly increased compared with that in the normal group(P<0.01).Compared with the model group,the p38 protein activity in Zusanli group and Yinglingquan group was significantly reduced(P<0.05).In the hypothalamus:compared with the Yingu group,the p38 of Zusanli group was significantly reduced(P<0.05).In the spinal cord:compared with Yinlingquan group and Yingu group,Zusanli group was significantly decreased(P<0.05).Conclusion Electroacupuncture on Zusanli point and Yinlingquan point both can improve the AWR score of FGIDs rats and reduce their visceral sensitivity,which may be related to the inhibition of p38 MAPK protein activation by electroacupuncture.Moreover,the regulation effect of electroacupuncture on p38 MAPK expression at Zusanli point is more obvious than that at yinlingquan point,which has acupoint specificity.
