DNA损伤修复基因XRCC1 rs25487单核苷酸多态性与男性不育关系的Meta分析

摘要 目的通过Meta分析系统评价DNA损伤修复基因XRCC1 rs25487基因多态性与男性不育的相关性。方法通过检索PubMed、CBM、中国知网及万方数据库中关于XRCC1 rs25487基因多态性与男性不育相关性的病例对照研究,由两名独立研究者按照纳入及排除标准获取文献,收集相关数据。采用Review Manager 5.3软件对纳入文献进行数据处理,通过5种比较模型分析XRCC1 rs25487多态性与男性不育发生之间的相关性。结果共纳入符合标准的7篇文献,包含病例组1579例和对照组1013例。Meta分析结果显示XRCC1 rs25487多态性与男性不育发生之间无明显相关性。5种比较模型中,等位基因模型A vs.G:OR=1.06,95%CI(0.83,1.36),P=0.64;显性模型AA+AG vs.GG:OR=1.08,95%CI(0.76,1.54),P=0.67;隐性模型AA vs.GG+AG:OR=1.03,95%CI(0.77,1.36),P=0.85;纯合模型AA vs.GG:OR=1.03,95%CI(0.77,1.36),P=0.85;杂合模型AG vs.GG:OR=1.04,95%CI(0.72,1.51),P=0.82。以种族和病例数(>200或<200)进行亚组分析,结果显示5种模型下差异均无统计学意义(P>0.05)。结论DNA损伤修复基因XRCC1 rs25487位点多态性与男性不育发生之间无明显相关性。 Objective:To evaluate the correlation between the DNA damage repair gene XRCC1 rs25487 gene polymorphism and male infertility through a meta-analysis.Methods:The case-control studies on the correlation between the XRCC1 rs25487 gene polymorphism and male infertility were searched from PubMed,CBM,CNKI and WANGFANG databases.The relevant data and literatures were obtained and collected according to the inclusion and exclusion criteria by two independent researchers.Review Manager 5.3 software was used to process the included literatures,and analyze the correlation between XRCC1 rs25487 polymorphism and male infertility through five comparative models.Results:Seven papers met the inclusion criteria,including 1579 cases in the case group and 1013 cases in the control group.Meta analysis revealed that no correlation between XRCC1 rs25487 polymorphism and male infertility.Allele model A vs.G:OR=1.06,95%CI(0.83,1.36),P=0.64;Dominant model AA+AG vs.GG:OR=1.08,95%CI(0.76,1.54),P=0.67;Recessive model AA vs.GG+AG:OR=1.03,95%CI(0.77,1.36),P=0.85:Homozygous model AA vs.GG:OR=1.03,95%CI(0.77,1.36),P=0.85;Heterozygous model AG vs.GG:OR=1.04,95%CI(0.72,1.51),P=0.82.Subgroup analysis was conducted based on race and number of cases(>200 or<200).The results showed that there was no significant difference among the five models(P>0.05).Conclusions:There is no significant correlation between the DNA damage repair gene XRCC1 rs25487 polymorphism and male infertility.
