摘要
根据前期研究及实验室现有条件,通过对磷脂酰胆碱物化性质的分析,研究了从鲤鱼下脚料中提取磷脂酰胆碱的复合酶法工艺。从单因素试验中可知,中性蛋白酶的最适添加量为0.03%,最佳酶解时间为60 min,最佳酶解温度为45℃,最佳酶解pH为7.5。eDTA溶液对酶的激活作用相对比NaCl溶液并不明显,所以选择氯化钠溶液作为激活剂。通过正交试验得到了影响提取磷脂酰胆碱的主次因素,中性蛋白酶对试验影响最大,木瓜蛋白酶次之,pH对酶活性有较大影响,激活剂增加了酶解效率,而胰蛋白酶对试验影响不大,进而确定了最佳的复合酶法提取工艺组合:木瓜蛋白酶添加量0.04%、中性蛋白酶添加量0.03%、胰蛋白酶添加量0.0175%、NaCl溶液0.20%、pH 7.5。
According to previous research and existing laboratory conditions,a compound enzymatic process for extracting phosphatidylcholine from carp waste was studied through analyzing physicochemical properties of phosphatidylcholine.In single factor experiment,the optimum amount of neutral protease was 0.03%,the optimum time for enzymatic hydrolysis was 60 min,the optimum temperature of enzymatic hydrolysis was 45℃,and the optimum enzymatic hydrolysis pH was 7.5.The activation of enzymes by EDTA solution was less obvious than that by NaCl solution.The primary and secondary factors affecting the extraction of phosphatidylcholine were obtained by orthogonal test.Neutral protease had the greatest influence,and then papain on the experiment.pH had a great influence on enzyme activity.And activators increase enzymatic hydrolysis efficiency.However,trypsinase had little effect on the experiment,and then the optimum combination of enzymatic extraction technology was determined.The addition of neutral protease was 0.03%,the amount of trypsin added was 0.0175%,NaCl solution is 0.20%,and pH was 7.5.
