摘要
目的研究散结通脉方对ApoE^-/-小鼠动脉粥样硬化模型血清中炎症因子含量及主动脉根部Orai1、STIM1蛋白表达的影响。方法采用高脂饲料(2%胆固醇、0.4%胆酸盐、10%脂肪、69.6%普通饲料)喂养ApoE^-/-小鼠12周建立动脉粥样硬化斑块模型,造模同时灌胃给予供试药品混悬液治疗。给药周期结束,检测各组动物血清TC、TG、HDL-cho、LDL-cho含量及TC/HDL-cho比值;血清hs-CRP、E-selectin、IL-6含量;主动脉根部STIM1、Orai1蛋白表达。结果散结通脉方13.5、6.75和3.40 g/kg 3个给药组具有明显降低模型动物血清TC、TG和LDL-cho的作用,与动脉粥样硬化模型组比较,分别为P<0.01或P<0.05;13.5 g/kg剂量组血清E-selectin、IL-6水平明显低于模型组,具有统计学差异(P<0.05);13.5、6.75 g/kg 2个剂量组主动脉根部Orai1、STIM1蛋白表达及血清中hs-CRP水平均明显低于模型组,具有统计学差异(P<0.01或P<0.05);3.40 g/kg剂量组主动脉根部Orai1蛋白表达明显低于模型组,具有统计学差异(P<0.05)。结论散结通脉方可以通过下调钙离子信号蛋白的表达抑制血管平滑肌细胞增殖迁移,并通过发挥调脂、抗炎等多种效应,对动脉粥样硬化发生发展过程中多因素多环节起到调节作用,进而延缓甚或逆转不稳定斑块的病理进程。
Objective To study the effect of Sanji Tongmai decoction on the content of inflammatory factors in serum and the expression of Orai1 and STIM1 proteins in aortic root in atherosclerosis model of ApoE-/-mice.Methods ApoE-/-mice were fed with high-fat diet (2% cholesterol,0.4% cholate,10% fat,69.6% common diet) for 12 weeks to establish the atherosclerotic plaque model.At the end of the drug administration cycle,the contents of TC,TG,HDL-cho,LDL-cho and the ratio of TC/HDL-cho in the serum of each group were detected.Serum contents of hs-CRP,E-selectin and IL-6;Expression of STIM1 and Orai1 proteins in aortic root.Results Sanji Tongmai decoction 13.5 g/kg,6.75 g/kg and 3.40 g/kg had the effect of significantly reducing the contents of serum TC,TG and HDL-cho in the model animals,which were P<0.01 or P<0.05,respectively,compared with the AS model group.The serum E-selectin and IL-6 levels of the 13.5 g/kg group were significantly lower than those of the model group,with statistical differences (P<0.05).The expression of Orai1 and STIM1 proteins and serum hs-CRP levels in the aortic root of the two dose groups were significantly lower than those in the model group (13.5 g/kg and 6.75 g/kg),with statistical differences (P<0.01 or P<0.05).The expression of Orai1 protein in the aortic root of the 3.40 g/kg group was significantly lower than that in the model group,with a statistical difference (P<0.05).Conclusion Sanji Tongmai decoction can inhibit the proliferation and migration of vascular smooth muscle cells by down-regulating the expression of calcium ion signaling protein,and play a role in regulating multiple factors and links in the development of atherosclerosis by exerting various effects such as lipid regulation and anti-inflammatory,thus delaying or even reversing the pathological process of unstable plaques.
