长链非编码RNASNHG5在重度子痫前期患者胎盘中的表达及其通过miR-155/CXCR4对滋养细胞侵袭的影响 被引量:6

摘要 目的观察长链非编码RNA(lncRNA)小核仁RNA宿主基因5(SNHG5)在重度子痫前期(sPE)患者胎盘组织中的表达,及其通过调控微小RNA 155(miR-155)对人滋养细胞(HTR8)侵袭功能的影响。方法qRT-PCR检测30例重度子痫前期(sPE组)与30例正常产妇(正常组)胎盘组织中lncRNA-SNHG5的表达水平。培养人滋养细胞(HTR8),分为:过表达组、抑制组和对照组(NC组),分别转染过表达质粒(pcDNA3.1-SNHG5)、siRNA-SNHG5和空质粒(pcDNA3.1)。Transwell法检测各组HTR8细胞侵袭能力。qRT-PCR、Western-blot检测各组中lncRNA-SNHG5、miR-155及其靶基因特异性趋化因子受体4(CXC chemokine receptor 4,CXCR4)的表达。结果qRT-PCR结果显示,与正常组相比,sPE组胎盘组织中lncRNA-SNHG5的表达显著下降(P<0.05)。与对照组相比,lncRNA-SNHG5在过表达组水平升高,在抑制组水平降低(P<0.05);Transwell结果显示,与对照组相比,过表达组穿过小室细胞数量升高,抑制组穿过小室细胞数量降低,差异有统计学意义(P<0.05);过表达组miR-155在mRNA水平低于抑制组(P<0.05),过表达组CXCR4在mRNA和蛋白水平均高于抑制组(P<0.05)。结论子痫前期患者胎盘中IncRNA-SNHG5表达下降。IncRNA-SNHG5可能通过影响miR-155/CXCR4途径导致滋养细胞侵袭能力下降,可能是PE发病的潜在机制。 Objective To observe the expression of long non-coding RNA(lncRNA)small nucleolar RNA host gene 5(SNHG5)in placenta tissues of severe preeclampsia,and its effect on the invasion ability of human normal trophoblast(HTR8)by regulating microRNA 155(miR-155).Methods The qRT-PCR was used to detect the expression of lncRNA-SNHG5 in placental tissues of 30 cases of severe preeclampsia(sPE group)and 30 cases of normal late pregnancy(normal group).Overexpression plasmids(pcDNA3.1-SNHG5),siRNA-SNHG5,and empty plasmids(pcDNA3.1)were transfected into HTR8 cells,named as overexpression group,siRNA-SNHG5 group and control group(NC group).Transwell method was used to detect the invasion ability of HTR8 cells in each group.The qRT-PCR and Western blot were used to detect the expression of lncRNA-SNHG5,miR-155 and its target gene CXCR4 in each group.Results Compared with normal group,the expression of lncRNA-SNHG5 in the placenta in sPE group was significantly reduced(P<0.05).The qRT-PCR showed that compared with control group,the level of lncRNA-SNHG5 was increased in overexpression group and decreased in siRNA-SNHG5 group(P<0.05).Transwell results showed that compared with control group,the number of cells passing through the Transwell chamber was increased in overexpression group,but decreased in siRNA-SNHG5 group(P<0.05).The mRNA level of miR-155 in overexpression group was lower than that in siRNA-SNHG5 group,and the mRNA and protein levels of CXCR4 in overexpression group were higher than those in siRNA-SNHG5 group(P<0.05).Conclusion The lncRNA-SNHG5 is decreased in the placenta of preeclampsia patients.The lncRNA-SNHG5 may lead to the decreased trophoblast invasion ability by affecting the mir-155/CXCR4 pathway,which is a potential mechanism of PE pathogenesis.
