IRAK-M基因SNP位点rs1821777多态性与成人支气管哮喘的相关性

摘要 目的:探讨IRAK-M基因SNP位点rs1821777多态性与成人支气管哮喘的相关性。方法:分别选取成人哮喘患者541人作为哮喘组和健康体检者514人作为对照组,收集2组研究对象的性别、年龄、血常规检查、血清免疫球蛋白E(IgE)及肺功能测定结果[包括1 s用力呼气容积(FEV1)、FEV1实测值/预测值、用力肺活量用力肺活量(FVC)、FEV1/FVC];抽取2组患者空腹外周静脉血3 mL,采用SNaPshot方法检测IRAK-M基因SNP位点rs1821777多态性、采用卡方检验比较2组患者基因型及等位基因分布差异。结果:哮喘组患者外周血嗜酸性粒细胞计数、嗜酸性粒细胞比例及血清IgE(经过对数转换后)水平均高于对照组,FEV1实测值/预测值、FEV1/FVC则均低于对照组,差异均有统计学意义(P<0.05);2组研究对象基因型分布和等位基因频率的差异均无统计学意义(P>0.05);与CC组比较,TT+CT组哮喘患者FEV1实测值/预计值、FEV1/FVC%降低,外周血嗜酸性粒细胞比例、外周血免疫球蛋白水平增高(P<0.05)。结论:IRAK-M基因SNP位点rs1821777多态性参与成人支气管哮喘临床表型特征的调节,但与成人哮喘易感性无关。 Objective:To investgate the correlation between the rs1821777 polymorphism in IRAK-M gene SNP and adult bronchial asthma.Methods:541 adult patients with asthma(as Asthma group)and 514 healthy controls(as the control group)were selected.Detailed historical medical records were collected,and the lung function examination,blood routine examination and serum IgE examination of all the subjects were completed.The SNaPshot was used to detect the rs1821777 polymorphism of the IRAK-M gene SNP locus in peripheral blood.Chi-square test was used to compare the distribution difference of rs1821777 polymorphism between Asthma group and the control group.Results:There was not statistically significant difference in the distribution of CC,CT,TT genotypes between the rs1821777 locus in Asthma group and the control group(P>0.05).Compared with CC group,the measured/predicted value of FEV1 and FEV1/FVC%in asthma patients with rs1821777 T genes were lower,and the ratio of peripheral blood eosinophils and IgE level were higher.The difference was statistically significant(P<0.05).Conclusion:The IRAK-M gene SNP lo cus rs1821777 polymorphism is involved in the regulation of clinical phenotype characteristics of adult patients with asthma,but was not related to adult asthma susceptibility.
