摘要
目的:通过观察调神畅情针法对PPD(帕金森抑郁)模型大鼠血清及脑组织中Ghrelin(胃促生长素)、NPY(神经肽酪氨酸)含量表达变化的影响,探讨调神畅情针法对帕金森病抑郁模型大鼠的作用机制。方法:采用6-OHDA纹状体双靶点注射法及慢性不可预见性温和应激法(CUMS)制备PPD大鼠模型,按随机数字表法分为5组,正常组、假手术组、模型组、药物组、调神畅情针法组,每组10只。采用不同的干预方法,干预28 d后,通过ELISA检测大鼠血清及脑组织中Ghrelin、NPY含量。结果:模型组大鼠血清及脑组织中Ghrelin、NPY含量均下降,与正常组、假手术组相比有显著差异(P<0.01);调神畅情组大鼠血清及脑组织中Ghrelin、NPY含量均有上升,与模型组相比有显著差异(P<0.01);西药组与模型组相比大鼠血清及脑组织中Ghrelin、NPY含量无显著差异(P>0.05)。结论:调神畅情针法有效治疗PDD的机制之一可能是提高PDD大鼠Ghrelin、NPY含量。
Objective:To observe the effects of Tiaoshen Changqing Acupuncture on ghrelin and NPY changes in serum and brain tissue of PPD(Parkinson’s depression)model rats,and explore the mechanism.Methods:PD model was established by 6-OHDA double target injection in striatum,and PPD rat model was prepared by chronic unpredictable mild stress(cums).According to the random number table method,the rats were divided into 5 groups:normal group,sham operation group,model group,drug group and Tiaoshen Changqing Acupuncture group,with 10 rats in each group.After 28 days of intervention,the contents of ghrelin and NPY in serum and brain tissue were detected by ELISA.Results:The contents of ghrelin and NPY in serum and brain tissue of model group were significantly decreased compared with normal group and sham operation group(P<0.01);the contents of ghrelin and NPY in serum and brain tissue of Tiaoshen Changqing Acupuncture group were increased,the differences between Acupuncture group and model group were significant(P<0.01);the contents of ghrelin and NPY in serum and brain tissue of rats in Western medicine group had no significant difference compared with model group(P>0.05).Conclusion:Increasing the content of ghrelin and NPY may be one of the mechanisms that Tiaoxiang Changqing Acupuncture can effectively treat PDD.
