摘要
目的:动态观测电针运动区对大脑中动脉闭塞(MCAO)大鼠海马内巢蛋白(Nestin)和神经生长因子β(NGFβ)的影响,探讨头针治疗缺血性中风的神经再生机制。方法:用线栓法制作大鼠MCAO模型,随机分为假手术组、模型组和针刺组。横木行走实验检测神经功能障碍的改变;Western blot检测Nestin的表达;实时定量RT-PCR方法检测海马组织NGF mRNA基因表达变化。结果:随治疗时间的延长针刺组横木行走实验评分呈升高趋势,在第14天与模型组比较差异具有统计学意义(P<0.01);不同时间点针刺组大鼠海马巢蛋白表达与模型组比较有显著提高(P<0.01);与模型组比较,针刺组大鼠NGF mRNA基因表达显著提高(P<0.01)。结论:电针运动区能显著上调大鼠海马组织Nestin蛋白及NGF mRNA基因表达,促进神经再生,加快功能恢复。
Objective:To observe the effect of electro-needling motor areas on NGF-β(nerve growth factor beta)and Nestin in MCAO(middle cerebral artery occlusion)rats,and to explore the mechanism of scalp acupuncture in treating ischemic stroke.Methods:The rats were randomly divided into the sham operation group,the model group and the EA group.MCAO model were established by the suture-occluded method in rats.The scoring of beam walking test was used to measure the dysneuria.The expression of Nestin was detected by Western blot method,and the expression of NGF mRNA was determined by RT-PCR.Results:The scores of beam walking test showed increasing trend as treating time extended in the EA group;of which there was significant difference between the EA group and the model group on 14th day(P<0.01).The Nestin expressions were significant higher in the EA group than those in the model group at different time points(P<0.01).Compared to the model group,the expression of NGF mRAN was significantly increased in the EA group(P<0.01).Conclusion:Electro-needling motor areas can obviously up-regulate the expressions of Nestin and NGF mRNA in hippocampus,promote the regeneration of cranial nerve and speed up the recovery of nerve function of MCAO rats.
